【摘要】 [目的]研究人肺腺癌GLC鄄82细胞中不同HSP70表达水平对该细胞放、化疗敏感性的影响。[方法]检测GLC鄄82细胞热处理前后HSP70表达水平的变化,以不同时间点确定为HSP70正常表达组、增强表达组、恢复表达组,分别以化疗、放疗、化疗结合放疗等方法处理,用MTT法测各组细胞存活率,比较HSP70不同表达水平对放化疗敏感性的影响。[结果]GLC鄄82细胞热休克处理前后HSP70表达水平明显不同,热休克处理后12h细胞HSP70表达最高,48h后达到预热前水平;各组细胞经不同剂量放射线作用后采用单靶多击模型拟和细胞生存曲线,显示HSP70正常表达组和HSP70恢复表达组生存曲线基本重合,增强表达组生存曲线右上移位,肩区增宽;不同化疗药物对HSP70增强表达组的抑制率较其它两组低,差异有统计学意义,虽然放化疗联合能显著提高对同一HSP70表达水平的GLC鄄82细胞的杀伤效果,但是HSP70增强表达组细胞对放化疗的联合杀伤敏感性较其它组低,差异有统计学意义。[结论]HSP70增强表达可以降低GLC鄄82细胞对放疗、化疗及放化疗联合作用的敏感性。更多还原

【Abstract】 To investe the compact of HSP70 expression and sensitivity of radiotherapy and chemotherapy on pulmonary carcinoma GLC-82 cells.Expression of heat-treated GLC-82 cells were examined.Groups of normal,The levels of expression were dertermined at various time point,with radiotherapy,chemotherapy or radio-chemotherapy separately.Survival rates of cells were measred with MTT methods.The levels of expression of GLC-82 cells were significantly different before and after heat treatment,with the peak expression at 12 hours,restored to control level at 48 hours after heat treatment.The cell survival curves were basically similar in groups of normal and restored expression after various dosages of radiotherapy,while the curve was shifting up and right in group of over-expression.The inhibit rate was significantly lower in group of over-expression than other two groups after different dosages of chemotherapy.The killing effects of GLC-82 cells were markedly increased in group of normal and restored expression after combined therapy,but not in group of over-expression.[Conclusion]Over expression of HSP70 may decrease the sensitivity to radiotherapy,chemotherapy and both combination for GLC-82 cells.更多还原
