尿激酶型纤溶酶原激活物及其抑制物-1表达与脑微血管改变的实验研究
The changes in brain microvasculature and the expression of uPA and PAI-1 in ischemic foci and perifocal areas after focal cerebral ischemia/reperfusion in rats.
【摘要】 目的探讨脑缺血再灌注病灶周围区脑微血管改变及与尿激酶型纤溶酶原激活物(uPA)、纤溶酶原激活物抑制物1(PAI1)表达的关系。方法应用光镜、电镜、免疫组织化学、原位分子杂交等技术,观察局灶脑缺血2h再灌注1、7、14、21d病灶周围区脑微血管结构改变及uPA、PAI1表达情况。结果脑缺血2h再灌注7、14d病灶周围区见新生微血管内皮细胞,且uPA、PAI1表达明显增加,uPA阳性单位7、14d分别为6.8±3.9、5.8±2.1,PAI1阳性单位7、14d分别为12.8±2.6、10.7±3.1。结论脑缺血再灌注后期病灶周围区有脑微血管内皮细胞再生,uPA、PAI1可能参与了微血管重建。
【Abstract】 Objective To study the changes in brain microvasculature in peripheral zones of cerebral ischemia/reperfusion and the expression of uPA and PAI-1. Methods By immunohistochemical staining, in situ hybridization, optical microscopy and electronic microscopy, changes in microvasculature and the expression of uPA and PAI-1 were observed in perifocal areas on the 1st, 7th,14th and 21st day after focal cerebral ischemia with reperfusion. Results Neogenetic endothelial cells in capillary appeared and expression of uPA, PAI-1 increased significantly on the 7th and 14th day after cerebral ischemia with reperfusion. The positive unit of uPA in reperfusion areas was 6.8±3.9, 5.8±2.1 respectively on the 7th and 14th day, the positive unit of PAI-1 was 12.8±2.6, 10.7±3.1 respectively on the 7th and 14th day. Conclusions At late stage of cerebral ischemia/reperfusion, endothelial cells of brain microvascular regenerated, and uPA and PAI-1 may contributed to the reconstruction of microvasculature.
