逆转肿瘤细胞多药耐药性的四物合剂有效组分

【摘要】 目的:研究四物合剂逆转人红白血病细胞株K562/ADM多药耐药作用的药效活性组分。方法:用大孔吸附树脂分离四物合剂,并用体外细胞模型作药效评价,寻找发现活性部位,再用HPLCMS方法研究活性部位的化学组成,进而确定其主要活性组分。结果:熟地黄粗多糖对K562/ADM的耐药性有明显的逆转作用,其逆转倍数与对照组(无逆转剂)相比有显著差异(P<0.01),并且与主要活性部位的逆转作用无明显差异(P>0.05)。结论:熟地黄粗多糖能逆转K562/ADM细胞的多药耐药性,是四物合剂逆转K562/ADM多药耐药性的主要活性组分之一。

【Abstract】 Objective: To study the main active constituents of Siwu mixture for reversing the multidrug resistance (MDR) of human erythrocyte leukemic cell line K562/ADM. Method: Macroporous resin was used to separate Siwu mixture and an in vitro model was used to evaluate the reversal index of each fraction for discovery of active fraction. HPLC-MS was used for investigating the main active constituents of Siwu mixture. Result: The reversal index of the gross polysaccharide of Rehmannia glutinosa was significantly higher than that of blank control ( P <0.01). There was little difference between the reversal effect of the gross polysaccharide of Rehmannia glutinosa and that of the active fraction A( P >0.05). Conclusion: The gross polysaccharide of Rehmannia glutinosa is one of the main active constituents of Siwu mixture.
