高效毛细管气相色谱法测定片剂中丙戊酸钠含量
Capillary column gas chromatography determination of sodium valproate in sodium valproate tablets
【摘要】 目的建立高效毛细管气相色谱法测定丙戊酸钠片剂的含量。方法用水溶解丙戊酸钠,加盐酸使其转化为丙戊酸,用乙醚萃取净化样品。色谱柱为ATSE54高效毛细管色谱柱(15m×0.25mm,0.33μm)。柱前压0.015MPa;进样口温度215℃;检测器FID;温度215℃;载气为N2,流速18mL·min-1;空气150mL·min-1;氢气62mL·min-1;Makeup(氮气)10mL·min-1。结果丙戊酸在2~20g·L-1浓度范围内线性关系良好(r=0.9998),丙戊酸高、中、低(16.0,8.0,4.0g·L-1)回收率分别为100.3%,99.0%,98.8%。结论本法简便、快速、准确,可用于丙戊酸钠片的含量测定。
【Abstract】 OBJECTIVE To establish the content determination method of sodium valproate tablets by capillary column gas chromatography.METHODS Sodium valproate was inverted into valproic acid at pH<~4.5 ,and was extracted and purified with ether.Valproic acid was separated and indentified on capillary column (~15 m ×~0.25 mm ,~0.33 μm ) by gas chromatography.The column temperature:~180 ℃ ,split injection; injector temperature:~215 ℃ ; detector: FID,temperature:~215 ℃ ,carrier gas N_~2 ~18 mL ·min~-1 ; Air ~150 mL ·min~-1 ; H_~2 ~62 mL ·min~-1 ;Makeup(N_~2 )~10 mL ·min~-1 .RESULTS The calibration curve was linear within the concentration range of 2-~20 g ·L~-1 , r=~0.999 8 . The average recovery for valproic acid were ~100.3% ,~99.0% ,~98.8% .CONCLUSION The method is simple,rapid,accurate,and could be used for determination of sodium valproate tablets.
