新型超声造影剂全氟丙烷白蛋白微囊的制备
Preparation of a novel ultrasound contrast agent:;perfluoropropane-containing albumin microcapsules
【摘要】 目的制备全氟丙烷白蛋白微囊,并观察制剂稳定性及超声造影效果。方法采用超声-冷冻法制备全氟丙烷白蛋白微囊,库尔特粒度分析仪测定粒径分布,吹扫捕集-气相色谱-质谱组合分析仪测定全氟丙烷含量,并对其在实验犬体内的左心超声造影效果进行评价。结果制备的微囊平均粒径为(3.2±0.4)μm,88.1%的微囊粒径在2~7μm内,全氟丙烷含量(272.3±61.2)mg·L-1,在冷藏条件下保存6个月无变化。经静脉注射后可实现左心室腔显影增强,提高左室内膜边界识别。结论采用超声-冷冻法制备的全氟丙烷白蛋白微囊,外观圆整,粒径适宜,稳定性好,是一种理想的超声造影剂,可显著增强医学超声检测信号,改善超声图象质量。
【Abstract】 OBJECTIVE To prepare perfluoropropane-containing albumin microcapsules, and study its stability and efficacy. METHODS Perfluoropropane-containing albumin microcapsules were prepared by a ultrasonication-lyophilization method. The particle size distribution was determined by Coulter, and perfluoropropane was analyzed by using a system combining Tekmar 3 000 Purge and Trap Concerntrator with HP5972GC/MSD. The contrast effect of microcapsules in dogs left ventricle was studied. RESULTS The average size of microcapsales was (3.2±0.4)μm. The sizes of 88.1% microcapsules ranged from 2~7 μm. Perfluoropropane content was (272.3±61.2) mg·L-1. There were no significant difference in quality parameters between freshly prepared microcapsules and those stored for 6 months. The left ventricle opacification grades and endocardial border resolutionm were significantly improved after intravenous injection of the microcapsules. CONCLUSION The microcapsules prepared by ultrasonication-lyophilization method are stable. The microcapsules morphology is regular, and their size distribution is small and even. It can be used as an excellent ultrasound contrast agent which can enhance significantly the yield of ultrasound examination and thus allow better imaging.
