【摘要】 目的考察紫杉醇制备成脂质体后对其组织分布的影响。方法建立了大鼠组织中紫杉醇HPLC检测法。色谱条件为:流动相为甲醇-乙腈-水(4:3:3);色谱柱为Turner kromasil C18(4.6 mm×250 mm,5μn);检测波长为227 nm;炔诺酮为内标。结果此色谱条件下各组织的标准曲线、回收率等试验结果表明该方法符合生物样品的分析要求。大鼠静脉注射紫杉醇脂质体及紫杉醇注射液(10 mg·kg-1)后,紫杉醇脂质体在肝脏、脾脏和肺脏等脏器分布相对较多。结论大鼠给予紫杉醇脂质体及紫杉醇注射液后在各个脏器的分布量几乎均有极显著性差异。更多还原

【Abstract】 OBJECTIVE To investigate the tissue distribution of paclitaxel liposome in rats.METHODS The HPLC method was used to determine the drug concentration in various tissues. The HPLC method was performed on a Turner kromasil C18 column (4.6 mm × 250 mm, 5 μm) with the mobile phase of methanol-acetonitrile-water (4:3 : 3). Paclitaxel was determined by UV absorbance at 227 nm.The internal standard was norethisterone.RESULTS The HPLC method was fit for determining the tissue distribution of paclitaxel. Paclitaxel liposome was mainly distributed in liver, spleen and lung in rats after iv 10 mg· kg-1 . CONCLUSION There is significant difference between the tissue distributions of rats after paclitaxel liposome and paclitaxel injection.更多还原
