苦玄参HPLC指纹图谱研究

【摘要】 目的研究苦玄参的高效液相色谱指纹图谱,为科学评价及有效控制其质量提供可靠方法。方法采用HPLC-DAD分析法,梯度洗脱,测定了10批苦玄参样品。色谱条件为:YMC J’sphere ODS-M80(4.6 mm×150 mm,4 μm)分析柱,柱温为35℃; 流动相A为乙腈;流动相B为1%醋酸水溶液。流动相A梯度洗脱(15%-45%乙腈),分析时间为65 min,时间为0,5,10,60, 65 min,A(%)为15,15,20,45,5,流速为1.0 mL·min-1,检测波长为254 nm。结果10批苦玄参样品得到的指纹图谱有8个共有峰,通过与对照品的保留时间及紫外光谱比较,1,2,6,7,8号峰分别鉴定为顺式阿克苷、阿克苷、苦玄参苷IB苦玄参苷Ⅳ和苦玄参苷IA。结论建立的HPLC指纹图谱分析法能较好地反映苦玄参中三萜和苯乙醇苷两大药效部位,可用于苦玄参的质量控制。

【Abstract】 OBJECTIVE To establish a sensitive and specific HPLC method for the quality control of Picria fel-terrae Lour.METHODS HPLC method was applied for the quality assessment of Picria fel-terrae Lour. The preparation of sample, the HPLC column, mobile phase, elution mode (isocratic and gradient) and gradient program were optimized in order to obtain high quality HPLC profile. The HPLC system consisted of a Waters 2695 pump and a 996 photodiode-array detector (DAD). Data were acquired and processed with the Empower workstation. HPLC analysis was performed on a YMC J’sphere ODS-M80 (4.6 mm×150 mm,4 μm) with the mixture of acetonitrile (A) and 1% acetic acid aqueous solution (B) as the mobile phase in gradient mode. The concentrations of solvent A were 15% , 15% , 20% , 45% and 45% at 0, 5, 10, 60 and 65 min, respectively. The column temperature was set at 35 ℃, the flow rate was 1.0 mL·min-1, and the detection wavelength was at 254 nm. RESULTS The HPLC fingerprint of Picria fel-terrae, showing 8 characteristic peaks, was established from 10 lots of Picria fel-terrae. By comparison of the retention time and the on-line UV spectra of chemical standards, peak 1, 2, 6, 7 and 8 were identified as cis-acteoside, acteoside, picfeltarraenin IB, picfeltarraenin Ⅳ and picfeltarraenin IA, respectively. CONCLUSION The HPLC fingerprint of Picria fel-terrae with high specificity can be used to control its quality.
