RP-HPLC测定齐多夫定棕榈酸酯脂质体药物含量

【摘要】 目的建立测定齐多夫定棕榈酸酯脂质体中药物含量方法。方法采用BetasilODS柱(4.6mm×200mm,5μm);流动相为乙醇-水(85:15),流速1mL·min-1,检测波长266nm,柱温为室温。结果在本色谱条件下齐多夫定棕榈酸酯与辅料及溶剂峰分离度符合要求,在10.3-103μg·mL-1内线性关系良好(r=0.9999,n=5),此方法回收率在100.1%~101.0%,日内及日间RSD均小于2%(n=5)。结论方法准确可靠、简单快速,可用于齐多夫定棕榈酸酯脂质体含量测定

【Abstract】 OBJECTIVE To establish a RP-HPI,C method for the determination of zidovudine palmitate in liposomes. METHODS The separation was performed with a Betasil ODS eolumn(4.6 mm × 200 mm, 5 μm) at mom temperature, and the mobile phase consisted of ethanol-water(85: 15) .flow with the rate of 1 mL·min-1. Zidovudine palmitate was detected at 266 nm.RESULTS Zidovudine palmitate had a good linear relation over the range of 10.3 - 103 μg·mL1( r = 0.999 9, n = 5) .The RSDs of intra-day and inter-day variation were less than 2% . The average recovery was between 100.1 % - 101.0% . CONCLUSION This method is simple, accurate, sensitive and applicable for the determination of zidovudine palmitate in liposomes.
