竹红菌甲素诱导人黑色素瘤A375-S2细胞凋亡的分子机制
Study on the apoptotic molecule mechanism in A375-S2 cell induced by hypocrellin A
【摘要】 目的研究竹红菌甲素(hypocrellin A,HA)诱导人黑色素瘤(A375-s2)细胞死亡的分子生物学机制。方法应用结晶紫染色法、流式细胞技术及RT-PCR技术等。结果HA能够诱导A375-S2细胞产生凋亡,并诱导细胞周期停滞在S期,同时诱导细胞周期蛋白CyclinB1的mRNA表达增加。结论HA诱导细胞周期因子CylinB1 mRNA表达增加与细胞周期停滞在S-期密切相关,同时细胞周期的阻碍可能是诱导A375-S2细胞凋亡的一个重要原因。
【Abstract】 OBJECTIVE To investigate the apoptotic molecule mechanism in A375-S2 ceU induced by hypocrellin A.METHODS Crystal violet dying, Flow Cytometer technique and RT-PCR were operated.RESULTS HA induced A375-S2 cell to apoptosis and cell cycle was arrested in S-phase, at the same time, HA increased cyclinB1 mRNA expression.CONCLUSION The arresting of cell cycle in S-phase was tightly interrelated with the fact that HA induced the increase of cell cycle regulator CyclinB1 mRNA expression. The study showed that cell cycle arrest might be an important factor that HA induced A375-S2 cell to apoptosis.
