大孔吸附树脂对淫羊藿总黄酮的分离纯化工艺研究

【摘要】 目的 研究大孔树脂分离纯化淫羊藿总黄酮的工艺条件及树脂前处理的方法。方法 以淫羊藿总黄酮洗脱率和纯度为考察指标 ,确定DM130型大孔吸附树脂分离纯化淫羊藿总黄酮的吸附性能和洗脱参数 ,建立用紫外分光光度法进行大孔树脂前处理的方法。结果 DM130型大孔吸附树脂吸附容量以干树脂计为 12 0 2mg·g-1,用纯化水和不同浓度的乙醇依次洗脱 ,以 6 0 %乙醇洗脱效果最佳 ,洗脱率达 92 0 3% ,总干燥物中淫羊藿总黄酮含量达 75 39%。结论 大孔吸附树脂对淫羊藿总黄酮有较好的分离纯化作用 ,且工艺简单 ,成本低 ,易于工业化生产。

【Abstract】 OBJECTIVE To study the purification process of total flavones from Herba Epimedii with DM130 macroreticular resin and the method to protreat macroreticular resin METHODS The eluting power and purified ratio of total flavones were worked as an index The adsorbility and eluting parameters of the process were studied RESULTS The adsorption capacity of macroreticular resin was 12 02 mg·g -1 60% Ethanol was optimum mobile phase After being adsorbed and purified by macroreticular resin, the eluting ratio and purity of total flavones was 92 03% and 75 39%, respectively CONCLUSION Applying macroreticular resin to adsorb and purify total flavones is feasible
