α干扰素对TNF-α诱导宫颈癌Hela细胞凋亡及NF-κB表达的研究
Effect of IFN-α on apoptosis of cervical cancer Hela cells induced by TNF-α and NF-κB expression
【摘要】 目的:探讨应用α干扰素与宫颈癌细胞核因子κΒ(NF-κB)表达的关系以及对肿瘤细胞凋亡的影响。方法:采用TUNEL法检测细胞凋亡,Westernblot分析方法检测NF-κB表达。结果:100U/ml的α干扰素与10ng/ml肿瘤坏死因子α(TNF-α)联合作用24h后,细胞凋亡率达到92%;与15ng/mlTNF-α联合作用12h后,凋亡率上升至92.6%。α干扰素不影响NF-κB的表达,TNF-α可显著增加NF-kB的表达(P<0.01),α干扰素预先作用后加用TNF-α可以显著抑制NF-κB的表达(P<0.01)。结论:α干扰素能够显著抑制TNF-α诱导宫颈癌Hela细胞NF-κB的表达,增强TNF-α诱导Hela细胞凋亡的作用。
【Abstract】 Objective: To explore the effects of interferon-α (IFN-α) on expression of nuclear factor (NF) -κB in cervical cancer cells and the apoptosis. Methods: Apoptosis was determined by using TUNEL method and the expression of NF-κB was analyzed by using Western blot. Results: After incubation with 100 U/ml of INF-α and 15 ng/ml of tumor necrosis factor-α (TNF-α) for 24 hours, the rate of cells apoptosis was 92%. After incubation with 15 ng/ml of TNF-α for 12 hours, the rate of apoptosis was 92.6%. Interferon-α didn’t affect the expression of NF-κB,while TNF-α could increase the expression of NF-κB(P<0.01). Pretreatment of INF-α and then TNF-α could inhibit the expression of NF-κB significantly(P<0.01). Conclusion: Interferon-α could suppress the activation of NF-κB of Hela cells and enhance apoptosis induced by TNF-α in Hela cells.
