头孢噻肟和他唑巴坦的体内抗菌作用研究

【摘要】 目的::对头孢噻肟及头孢噻肟和他唑巴坦合剂的不同配比制剂对细菌腹腔感染小鼠的治疗进行研究,寻找最佳配方。方法:将健康小鼠随机分组,建立产超广谱β内酰胺酶(ESBLs)埃希大肠杆菌和肺炎克雷伯杆菌、产β内酰胺酶金黄色葡萄球菌腹腔感染模型,感染1h后分别给予头孢噻肟和他唑巴坦1∶1, 3∶1, 5∶1, 7∶1不同配比制剂及头孢噻肟、他唑巴坦单剂,观察记录感染后7d小鼠的生存率。结果:头孢噻肟和他唑巴坦合剂(1∶1, 3∶1, 5∶1, 7∶1)对产ESBLs肺炎克雷伯杆菌、大肠杆菌腹腔感染小鼠的保护作用明显优于头孢噻肟单剂,半数有效剂时(ED50 )比较P<0. 01;头孢噻肟和他唑巴坦(1∶1, 3∶1)合剂对金黄色葡萄球菌产酶耐药菌腹腔感染小鼠的治疗作用明显优于头孢噻肟单剂,ED50比较P<0. 01。头孢噻肟和他唑巴坦(5∶1, 7∶1)治疗作用与头孢噻肟单剂相比无明显差异,ED50比较P>0. 05。结论:头孢噻肟与他唑巴坦合剂(1∶1, 3∶1)对肺炎克雷伯菌、大肠杆菌产酶耐药菌、产β内酰胺酶金黄色葡萄球菌腹腔感染的小鼠有显著的抗菌治疗作用,为较好的配方组合。

【Abstract】 Objective: To investigate the therapeutic effect on infecting peritonitis of mice with different formula of cefotaxime and tazobactam.Methods:The healthy mice were randomized into groups infected with extended-spectrum β-lactamases(ESBLs)-producing Escherichia.coli, K.pneumoniae,β-lactamases-producing S.aureus to establish peritonitis models. The mice separately recieved cefotaxime and tazobactam (1∶1,3∶1,5∶1,7∶1),cefotaxime alone,and tazobactam alone 1 hour after being infected.We observed the survival rate of mice after 7 days.Results:The median effective dose(ED 50) of cefotaxime and tazobactam (1∶1,3∶1,5∶1,7∶1) on intraperitoneal infections induced by ESBLs-producing K.pneumoniae and Escherichia.coli in mice were significantly lower than those of cefotaxime alone(P<0.01).The therapeutic effect of cefotaxime and tazobactam (1∶1,3∶1) on intraperitoneal infections induced by β-lactamases-producing S.aureus was better than that of cefotaxime alone(P<0.01). The ED 50 of cefotaxime and tazobactam (5∶1,7∶1 ) on intraperitoneal infections showed no significant difference with that of cefotaxime alone(P>0.05). Conclusion:The cefotaxime and tazobactam with 1∶1 and 3∶1 ratio have the best antibacterial activities against ESBLs-producing antibiotic K.pneumoniae ,Escherichia.coli, and β-lactamases-producing S.aureus.
