北京市社区人群胆固醇调节元件结合蛋白-1a基因多态性与血脂水平的关系

【摘要】 目的 了解SREBP -1a基因-3 6delG多态性在中国汉族人群中的分布情况及其与高脂血症的关系。方法 本研究以北京市西城区厂桥社区人群筛检出的、未经药物治疗的3 2 2例高脂血症患者、5 1例临界高脂血症患者及164名血脂正常者为研究对象,运用聚合酶链反应-限制性片段长度多态性(PCR -RFLP)检测SREBP -1a基因-3 6delG位点多态性。结果 SREBP -1a基因-3 6delG等位基因频率在血脂正常组、临界高脂血症组和高脂血症中分别为9 5 .%、11. 8%和10 . 3 % ,差异无统计学意义(P >0 . 0 5 )。血脂正常组和临界高脂血症患者中血清TC、LDL -C、HDL -C、TG、ApoAI和ApoB水平在不同基因型间差异无统计学意义;在高脂血症患者中,女性-3 6delG携带者血清TC水平低于G + /G +基因型,差异有统计学意义(P <0 . 0 5 ) ,而其余血脂指标在不同基因型间差异无统计学意义(P >0. 0 5 )。结论 SREBP -1a的G + /G +基因型可能与女性高脂血症患者血清TC水平的升高有关。

【Abstract】 Objective To determine the e ffects of Sterol Regulatory Element Bin ding Protein-1a (SREBP-1a) gene polymorphisms on the serum lipid levels in the m iddle-aged and elder people of China. Methods A total of 537 unrelated individ uals, aged 35 to 81, were selected from Xicheng District of B eijing. For all the subjects the serum lipids profiles were measured and the SR EBP-1a polymorphisms were analyzed (PCR- RFLP). According to serum TG and TC le vels, all subjects were classified into three groups: normolipidemic group (n =164), preclinical hyperlipidemic group (n=51) and hyperlipidemic group ( n=322). Results The frequencies of - 36delG allele of SREBP-1a gene in normolipidemic group, preclinical hyperlipidemic group and hyperlipidemic group were 0.095, 0.118 a nd 0.103 respectively. There were no significant differences in genotype frequenc ies or allele frequencies of the polymorphism among the three groups (P>0.05) . Th e serum TC levels of the individuals carrying G- allele were significantly lower than that of the G+/G+ carriers in the female hyperlipidemic group (P<0.05). Conclusion G+/G+ genotype of SREBP-1a is significantly related to the increased concentrations of serum TC in female h yperlipidemic subjects.
