禽流感病毒A/chicken/Mudanjiang/0823/2000(H9N2)株NP及M1基因的克隆及序列分析
Cloning and sequencing of NP and M1 gene of avian influenza virus strain A/chicken/Mudanjiang /0823/2000(H9N2)
【摘要】 参照已发表的A型禽流感核蛋白 (NP)、基质蛋白 (M 1)基因序列及其基因组特性 ,设计合成了一条通用反转录引物和两对特异性引物 ,采用RT_PCR法 ,成功克隆了禽流感病毒国内分离株A /chicken /Mudanjiang / 0 82 3/ 2 0 0 0(H9N2 )株的NP、M 1基因。序列测定结果为NP基因cDNA全长 14 97bp ,编码 4 98个氨基酸。M 1基因cDNA全长 75 9bp ,编码 2 5 2个氨基酸。将其序列与数株A型流感病毒 (H9N2 )NP及M 1基因序列进行比较 ,NP基因同源性为 90 5 1%~ 98 4 6 % ,氨基酸序列同源性为 95 38%~ 98 5 9%。M1基因同源性为 90 78%~ 97 36 % ,氨基酸序列同源性为95 6 3%~ 99 6 0 %。
【Abstract】 A piece of universal reverse transcription primer and two pairs of specific primers were designed according to the M1 and NP gene of AIV.The NP and M1 gene of Avian Influenza Virus A /chicken /Mudanjiang/0823/2000(H9N2) strain were amplified by RT_PCR .NP gene is 1 497 bp in length,encoding 498 amino acids.M1 gene is 759 bp in length, encoding 252 amino acids.Comparing with other published NP and M1 cDNA sequences, It shows that the homology of NP gene is from 90.51 % to 98.46 % and the homology of amino acids is from 95.38 % to 98.59 %, the homology of M1 gene is from 90.78 % to 97.36 % and the homology of amino acids is from 95.63 % to 99.60 %.
