bc1-2和bax及NF-kB在白藜芦醇诱导肝癌细胞凋亡中的作用
The role of bc1-2 and bax as well as NF-kB on liver cancer cell apoptosis induced by resveratrol
【摘要】 目的探讨白藜芦醇诱导肝癌细胞凋亡的途径。方法白藜芦醇体外处理HepG2肝癌细胞24h后,以免疫组化检测凋亡调控基因bc1-2和bax及NF-kB的表达。结果白藜芦醇处理组HepG2细胞bc1-2的阳性积分和NF-kB的阳性细胞密度均明显低于对照组(P<0.01);而bax阳性积分明显高于对照组(P<0.01)。结论白藜芦醇能下调HepG2细胞bc1-2基因的表达,上调bax的表达,同时抑制NF-kB的活化,这可能是其诱导HepG2细胞凋亡的途径之一。
【Abstract】 Objective To explore the pathway of resveratrol inducing apoptosis of liver cancer cell.Methods After resveratrol treated HepG2 liver cancer cell in vitro for 24 hours,the apoptosis regulating genes such as bc1-2 and bax as well as NF-kB were detected by using immunohistochemistrical method.Results The positive score of bc1-2 and positive cell density of NF-kB in resveratrol treating group were significangly lower than that in control group (P<0.01).The positive score of bax in resveratrol treating group were obviously higher than that in control group (P<0.01).Conclusions Resveratrol could downregulate the expression of bc1-2 gene of HepG2 cell,increase the expression of bax gene,and inhibit the activity of NF-kB.It was possible pathway of resveratrol inducing apoptosis of HepG2 cells.
