普通冠状病毒229E株的分子流行病学分析

【摘要】 目的明确哈尔滨地区普通冠状病毒229E(HumanCoronavirus229E,HcoV229E)株的流行和变异情况及其与SARSCoV的异同,为进一步掌握该地区常见上呼吸道病毒的流行规律及预防,乃至疫苗的制备打下基础。方法利用RTPCR法对2003年上半年采集的部分发热病人血清及血细胞进行筛选,同时采用基因测序、序列分析等手段对扩增的HcoV229ENgene片断进行蛋白和基因分析。结果55例标本中HcoV229ERNA阳性病例5例,占909%;测序结果看出哈尔滨地区检出229E株N基因的序列与已公布的HcoV229ENgene(295802)序列完全相同,所测片断为HcoV229ENgene的部分序列。该基因与猪流行性腹泻病毒、犬冠状病毒、TGEV及猫感染性腹膜炎病毒有一定同源性;但与已公布的SARSCoVNgene序列比较,相似性小于1%。结论(1)该地区发热病人普通冠状病毒229E株阳性率为909%;(2)哈尔滨地区流行的HcoVN基因无变异发生;(3)HcoV229E与猪流行性腹泻病毒、犬冠状病毒、TGEV和猫感染性腹膜炎病毒有一定的同源性;(4)哈尔滨地区HcoV229ENgene与SARSCoVNgene相似性小于1%。

【Abstract】 Objective To know the epidemic condition and the variation of HcoV-229E in Harbin,to analyse the difference between HcoV-229E and SARS-CoV. Methods We adaped RT-PCR to detect HcoV-229E RNA,and we adapted gene sequencing method to get HcoV-229E N gene sequence.Results We detected 55 samples,and found 5 positive cases,the positive percent was 9.09%;The analysis of sequence showed that The sequence of Harbin was the same to HcoV-229E N gene which had been published,the gene segment was part of HcoV-229E N gene;The sequencing result had less 1% similarity with SARS-CoV N gene.Conclusions (1) The positive percent of HcoV-229E in Harbin was 9.09%,there was coincidence results through the RT-PCR method and blood serum epidemiology method;(2) The HcoV-229E in Harbin had no variation;(3) There were homology between HcoV-229E and porcine epidemic diarrhea virus,Canine coronavirus,Transmissible gastroenteritis virus,feline infectious peritonitis virus;(4) The similarity of HcoV-229E N gene in Harbin and SARS-CoV N gene was less than 1%,the result accords with correlated report.
