新穿心莲内酯对体外活化小鼠巨噬细胞的影响

【摘要】 目的:考察新穿心莲内酯对体外活化的小鼠巨噬细胞活性影响,探讨其免疫抗炎活性。方法:采用BALB/C小鼠骨髓细胞定向分化巨噬细胞,应用化学发光法考察新穿心莲内酯对佛波豆蔻酸乙酯(PMA)刺激的小鼠巨噬细胞呼吸爆发,alamarBlue染色法和Griess法测定新穿心莲内酯对LPS诱导的小鼠巨噬细胞增殖及NO合成的影响。结果:新穿心莲内酯在30~150μmol/L浓度范围内对刺激的巨噬细胞呼吸爆发有抑制作用,在11·25~120μmol/L内对活化的小鼠巨噬细胞增殖及NO释放有明显抑制作用。结论:新穿心莲内酯具有免疫抑制活性。

【Abstract】 AIM: To investigate the effect of neoandrographolide on ac tivated mouse macrophages. METHOD: Macrophages were derived fro m bone marrow of BALB/C mice. Respiratory burst of macrophage was measured by lu minol-dependent cellular chemiluminescence when macrophage was stimulated by Ph orbol-12-myristate-13-acetate (PMA). Proliferation of macrophage was measure d by alamarBlue assay and NO production was detected by Griess assay when macrophage was induced by lipopolysaccharide (LPS). RESULT: Neo andrographolide (11.25~120 μmol/L) suppressed the respiratory burst dose-dep endently stimulated by PMA. It also inhibited LPS (5 μg/L) induced proliferati on and nitric oxide production in mouse macrophages. However, neoandrographolide at low concentrations (1.5~7.5 μmol/L) stimulated respiratory burst, prolif eration and nitric oxide production in mouse macrophages. CONCLUSION: The current data suggested that neoandrographolide could suppress the acti vation of mouse macrophage in vitro.
