抗猪传染性胃肠炎病毒单克隆抗体杂交瘤细胞株的建立
Development of hybridomas secreting monoclonal antibody specific to transmissible gastroenteritis virus of pigs
【摘要】 应用杂交瘤技术获得了1株分泌抗猪传染性胃肠炎病毒(TGEV)单克隆抗体(McAb)杂交瘤细胞2D6。经检测,其分泌的抗体亚类为IgG3;杂交瘤染色体数为88;间接ELISA检测细胞培养上清液效价为1∶256,诱生腹水抗体效价达1∶6×104,与猪流行性腹泻病毒(PEDV)、猪轮状病毒(PRV)、猪致病性大肠埃希氏菌(Escherichia coli)以及禽传染性支气管炎病毒(IBV)抗原之间均无交叉反应;ELISA阻断试验表明,TGEV特异性阳性血清对TGEV-McAb有明显的阻断作用,该McAb所识别的抗原是TGEV所特有的抗原决定基。
【Abstract】 A hybridoma cell line producing monoclonal antibody against transmissible gastroenteritis virus of pigs was established by the hybridoma technique.The antibody,designated as 2D6,of subclass IgG3 was isolated.The number of chromosome of the hybridoma cell line was 88.The ELISA titers of ascite and cell supernatant was 1∶6×104 and 1∶256,respectively.No cross-reaction was observed between the McAb and PEDV,PRV,Escherichia coli and IBV.Specific TGEV positive serum inhibited the reaction of the McAb with TGEV competitively.The McAb was highly specific to the antigenic determinant of TGEV.
