水产品中创伤弧菌的快速检测与分离
Rapid Identification and Isolation of Vibrio Vulnificus in Aquatic Products
【摘要】 目的:对水产品中创伤弧菌的快速检测与分离技术进行研究。方法:蛤蜊肉经高速匀浆处理后,在含有多粘菌数B的碱性蛋白胨水中增菌培养(35~37℃,12~16h),然后用PCR技术检测样品。PCR检测的目的基因是创伤弧菌的溶细胞毒素(hemolysin/cytolysin)基因,扩增产物为222bp。对PCR结果呈阳性的样品做原位杂交实验,用碱磷酶标记的寡核苷酸探针VVAP来分离创伤弧菌。通过对购买于青岛丹东路小市场的蛤蜊进行检验,分离出了2株创伤弧菌。结果表明:应用PCR、原位杂交技术能快速检测和分离水产品中存在的创伤弧菌。
【Abstract】 Objective:The method of rapid identification and isolation of Vibrio vulnificus in aquatic products was studied. Methods:After homogenized,the content of clam was incubated in alkaline peptone water containing with polymyxin B at 35~37 ℃ for 12~16 h. Primers targeting then the Vibrio vulnificus hemolysin/cytolysin gene was identified with one set of samples. The expansion product was 222 bp. If the PCR result was positive,an alkaline phosphatase-labeled oligonucleotide DNA probe was used to isolate Vibrio vulnificus in samples. Results:The method was used to identify the Vibrio vulnificus in clams bought at Dangdong Road market in Qingdao and two Vibrio vulnificus strains were isolated and identified. Conlusion:PCR and In situ hybridization technology can rapidly identify and isolate Vibrio vulnificus in aquatic products.
