滋补脾阴方药对大鼠脊髓损伤后IL-1β、caspase-3表达及细胞凋亡的影响
The influence of piyin remedy on apoptosis and expression of IL-1β and caspase-3 after spinal cord injury in rats
【摘要】 目的:研究大鼠脊髓损伤后白细胞介素-1茁(Interleukin-1茁,IL-1茁)半胱氨酸天冬特异性蛋白酶(cysteinglas-partatespecificprotease,caspase)表达和细胞凋亡的规律及三者之间的关系、滋补脾阴方药对它们的影响。方法:将成年SD大鼠随机分为假手术组、损伤组和滋补脾阴方药组,于伤后不同时间点(1h、8h、24h、72h)处死,用免疫组化染色检测IL-1茁、caspase-3阳性细胞,TUNEL法标记凋亡细胞。结果:免疫组化结果显示假手术组仅有少量IL-1茁和caspase-3阳性细胞表达;损伤组的两者均在8h表达最多,24h减少,72h减少至略多于假手术组。TUNET标记阳性细胞的时间分布特点与IL-1茁和caspase-3相似,8h最多,72h减少至较低水平。IL-1茁、caspase-3表达阳性细胞率和细胞凋亡指数三者间呈正相关;滋补脾阴方药使IL-1茁、caspase-3表达阳性细胞率和细胞凋亡指数降低。结论:大鼠脊髓损伤后,IL-1茁和caspase-3表达增强,凋亡细胞大量出现,三者间存在正相关;滋补脾阴方药能够抑制脊髓损伤后的炎症反应,减少细胞凋亡,减轻脊髓继发性损伤。
【Abstract】 Objective:To study the characteristic of apoptosis and the expression of interleukin-1β(IL-1β) and cysteingl aspartate specific protease(caspase),as well as relation among them and the influence of piyin remedy on them after spinal cord injury(SCI) in rats.Method:Adult SD rats were divided into three groups randomly veetebva shelf-operated controls and cord injury group and piyin remedy group, The rats were Killed at 1h 8h 24h and 72h after surgery The expression of IL-1β and caspase-3 was betermined by.immunohisto chemistry and the apoptosis was labeled by terminal deoxynuohisto transfersae-mediated dutp nick end labeling(TUNEL).Result:There was only a few cells that expressed IL-1β and decreased to a low level a litte higher than that of vertebra shelf-operated controls at 72h. The rates of TUNEL-positive cells were similar with that of IL-1β and caspase-3 after SCI in flood tide in 8h decresed to low level at 72h. There was positive correlation among expression of IL-β and expression of caspase-3 and apoptosis index.Piyin remedy reduced the expression of IL-1β and caspase-3 and the apoptosis index.Conclusion:Expression of IL-1β and caspase-3 increase and a large number of TUNEL-positive cells present after SCI in rats. There is positive correlation among expression of IL-1β and expession of casspase-3 and apoptosis index. Piyin remedy can suppress inflammatory reactim,reduce apotosis and lessen secondary injury of cord after SCI.
