光动力学治疗鲜红斑痣反应体系数学仿真的初步探讨

【摘要】 目的通过建立光动力学治疗鲜红斑痣中激光、光敏剂的分布及其相互作用关系的数学模型,对微血管中单态氧的产生过程进行仿真,分析各种因素对治疗的影响,对数学建模在光动力反应体系中的应用价值进行初步探讨。方法用MonteCarlo方法描述光在组织中的分布;用药代动力学描述光敏剂在血管中的变化规律;用Lambert-Beer定律和单态氧的量子产率来计算血管中的单态氧产量。结果单态氧在血管中的空间变化规律由光分布决定,血管中心产生少;时间变化规律由光敏剂的药代动力学决定,呈迅速衰减趋势。光敏剂的倍增比激光剂量的倍增产生更多的单态氧。直径大的血管平均单态氧量较直径小的血管少,难以被破坏。肤色深单态氧的产生少。结论 数学仿真能较好地描述光动力学治疗鲜红斑痣中各种因素对治疗的影响,数学建模是一种对光动力复杂作用体系的研究有独到之处的研究方法。

【Abstract】 Objective Through establishing mathematical model of the distribution of laser, photosensitizer and their interaction relationship, to simulate the production of singlet oxygen in vascular, analyze the effects of varied factors on photodynamic therapy of port wine stains, preliminarily explore the applicable value of mathematical modeling in photodynamic reaction system. Methods To formulate the distribution of laser by Monte Carlo model and photosensitizer by pharmacokinetics. To calculate the production of singlet oxygen in vascular by the law of Lambert-Beer and quantum yields of singlet oxygen. Results The spatial regularity of singlet oxygen is decided by the distribution of laser, and there is little singlet oxygen in the center of vessel. The temporal regularity of singlet oxygen is decided by pharmacokinetics of photosensitizer, which show that the tendency of singlet oxygen decreasing rapidly. Giving double amount of pholosensilizer would produce more singlet oxygen than giving double amount of laser. The vascular with big diameter has little amount of singlet oxygen than that with small diameter averagely and is difficult to be destroyed. There are little singlet oxygen in vessel with dark color. Conclusions Mathematical simulation is a good way to formulate the effects ol various factors on photodynamic therapy of port wine stains. Mathematical modeling is the particular method in studying photodynamic complex reaction system.
