花青素对人脐静脉内皮细胞基质金属蛋白酶影响

【摘要】 目的研究花青素对人脐静脉内皮细胞基质金属蛋白酶(MMP)1,2和9蛋白表达和活性的影响。方法分离人脐静脉内皮细胞,先用5mg/L人重组CD40配体(rCD40L)处理细胞24h,再分别用1,10,100μmol/L的花青素标准品矢车菊定3葡萄糖苷(C3G)和芍药定3葡萄糖苷(P3G)处理细胞24h。用WesternBlotting分别检测MMP1,MMP2,MMP9的蛋白表达量,用酶谱分析法检测MMP1,MMP2,MMP9的活性水平。结果花青素C3G和P3G组的MMP1,MMP2,MMP9的活力明显下降,且蛋白表达量也明显下降。结论花青素C3G和P3G能够降低rCD40L诱导的人脐静脉内皮细胞MMP1,MMP2,MMP9的蛋白表达和活性水平,对于增加动脉粥样斑块的稳定性有重要意义。

【Abstract】 Objective To explore the effect of anthocyanin on matrix metalloproteinases (MMP) expression and activity. Methods Human umbilical vein endothelial cell were isolated and incubated with 5μg/mL rCD40L for 24h. After that,the endothelial cell were incubated with 1μM,10μM,100μM C3G and P3G for another 24h, respectively. Methods of zymography and Western blotting were used to analyze MMP-1, MMP-2, MMP-9 activities and protein expression.Results Anthocyanin C3G and P3G inhibited MMP-1, MMP-2 and MMP-9 protein expression and their activities significantly.Conclusion Anthocyanin C3G and P3G inhibited MMP-1,MMP-2,MMP-9 protein expression and their activities.
