糖基化终产物对大鼠肾系膜细胞结缔组织生长因子作用的研究
Induction of connective tissue growth factor mRNA expression by advanced glycosylation end products in cultured rat renal mesangial cells
【摘要】 目的 :探讨糖基化终产物 (AGEs)对大鼠肾系膜细胞结缔组织生长因子 (CTGF)mRNA表达及细胞外基质 (ECM)合成的影响。方法 :在培养的大鼠肾系膜细胞中 ,分别加入不同浓度的糖化牛血清白蛋白 (AGE -BSA)及牛血清白蛋白 (BSA)进行刺激 ,ELISA法检测培养上清中的纤连蛋白 (FN)及Ⅳ型胶原 (ColⅣ )含量 ,RT -PCR检测细胞CTGFmRNA表达。结果 :与加入BSA组比较 ,AGE -BSA组CTGFmRNA表达明显增强 (P <0 . 0 1) ,上清中FN及ColⅣ合成增加 (P <0 . 0 1) ,与CTGFmRNA表达量之间呈正相关 (r=0 . 83)。结论 :AGEs能明显诱导大鼠肾系膜细胞CTGFmRNA的表达 ,提示AGEs可能通过上调CTGFmRNA的表达引起ECM积聚。
【Abstract】 AIM: To explore the effects of advanced gly cosyla tion end products (AGEs) on connective tissue growth factor (CTGF) mRNA expressi on and extracellular matrix (ECM) synthesis in cultured rat renal mesangial cel ls. METHODS: Rat renal mesangial cells were cultured in vitro un der standard conditions. The content of fibronectin (FN) and type Ⅳ collagen (ColⅣ ) were determined with ELISA after stimulation by AGE-BSA and BSA, respectively. CTGF mRNA expression was assessed by RT-PCR. RESULTS: CTGF mRNA was significantly increased by AGE-BSA in cul tured mesangial cells in a time-and dose-dependent manner compared with BSA cont rol (P<0.05). Synthesis of FN and Col Ⅳ was upregulated by AGE-BSA treatm ent in cultured rat mesangial cells (P<0.05), which was positively correlat e d with CTGF mRNA expression level. CONCLUSIONS: AGEs significantly upregulate CTGF mRNA expression in cultured mesangial cells, suggesting that AGEs induce ECM accumulation by up regulating CTGF mRNA expression.
