尖锐湿疣组织中的细胞凋亡与TRAIL受体的表达
Apoptosis and Expression of Tumor Necrosis Factor-related Apoptosis Inducing Ligand Receptors(TRAIL-R) in Lesions of Condyloma Acuminatum
【摘要】 目的 探讨尖锐湿疣患者皮损中肿瘤坏死因子相关凋亡诱导配体 (TRAIL)受体DR4,DR5的表达情况及其与细胞凋亡之间的相关性。方法 应用逆转录-聚合酶链反应 (RT PCR)检测 30例尖锐湿疣患者皮损中DR4,DR5mRNA的水平,并设 15例正常皮肤组织作对照;原位末端标记法(TUNEL)检测皮损中角质形成细胞的凋亡。结果 尖锐湿疣组织标本DR4,DR5mRNA的平均水平分别为 0. 98±0. 20, 1. 18±0. 16,与正常皮肤组织相比显著升高(P<0. 05);凋亡指数与DR4,DR5的水平有显著相关性(P<0. 05)。结论 DR4,DR5在尖锐湿疣组织中表达,并与病毒感染细胞的凋亡相关,可能在TRAIL/TRAIL R系统诱导病毒感染细胞凋亡中发挥重要作用。
【Abstract】 Objective To investigate the expression of tumor necrosis factor-related apoptosis-inducing ligand death receptor 4 (DR4) and death receptor 5 (DR5) mRNA in the lesions of condyloma acuminatum (CA) at the mRNA level and its correlation with keratinocyte apoptosis.Methods Total RNA isolated from the lesions of CA was reversely transcribed into the first strand cDNAs, and the products were amplificated by PCR. After electrophoresising and dying with EB, the expression of DR4 and DR5 mRNA were analyzed. Terminal deoxynucleotidyl transferase(TdT)-mediated biotin dUTP nick end-labeling(TUNEL) was used to evaluate apoptosis cells.Results In all 30 CA tissues the expression of DR5 and DR4 mRNA were stronger compared to the 15 non-CA foreskin tissues(P<0.05). Apoptosis correlated with expression of DR4 and DR5(P<0.05).Conclusion It implies that TRAIL apoptotic pathway is functional in human keratinocytes and it is likely to be mediated through DR4 and/or DR5.
