雌激素和孕激素及肝素结合表皮生长因子对子宫内膜癌细胞中整合素β3表达的调节

【摘要】 目的通过体外实验研究雌激素、孕激素和肝素结合表皮生长因子(HB-EGF)对子宫内膜癌细胞中子宫内膜容受性的标志整合素β3(integrinβ3)表达的调节,探讨整合素β3表达的意义及子宫内膜容受性的调节因素。方法体外培养高分化子宫内膜癌细胞Ishikawa,按实验目的分别加入雌激素、孕激素、雌孕激素联合、雌孕激素及孕酮抑制药(RU486)、HB-EGF刺激48h后,采用逆转录-聚合酶链反应(RT-PCR)、Westernblot、原位杂交、免疫细胞化学方法测定各个条件下内膜癌细胞中整合素β3mRNA及蛋白表达。结果雌激素单独以及雌激素、孕激素同时作用均使Ishikawa细胞中整合素β3mRNA及蛋白的表达显著下降;RU486可减弱雌激素、孕激素对integrinβ3的下调作用;而孕激素单独应用对整合素β3表达无明显影响;HB-EGF可显著提高整合素β3的表达。结论整合素β3mRNA及蛋白的表达受甾体激素和生长因子两方面的调节。

【Abstract】 Objective To study the regulation of the integrin β3 expression in a well differentiated endometrial adenocarcinoma cell line (Ishikawa) by estrogen (E2), progestin (P 4 ) and heparin-binding epidermal growth factor (HB-EGF). Methods Ishikawa cells were treated with nothing or with 17β-E2 (10 -8 mol·L -1 ), P 4 (10 -6 mol·L -1 ), HB-EGF (10 ng·mL -1 ), 17β-E2 plus P 4 , 17β-E2 plus P 4 and antiprogestin (RU 486 10 -5 mol·L -1 ) for 48 hours. The expression of the integrinβ3 was determined with reverse transcription-polymerase chain reaction ( RT-PCR), western bolt analysis, in situ hybridization, and immunocytochemistry. Results The expression of integrin β3 was significantly reduced by 17β-E2 treatment or by 17β-E2 plus P 4 treatment. The addition of mifepristone RU486 reversed the suppression of integrin β3. P 4 had no significant effect on integrin β3 expression. HB-EGF significantly increased the integrin β3 expression. Conclusion The expression of the imtegrin β3 is regulated by both steriod hormones and growth factors.
