黄芪提取物对犬心力衰竭模型的作用

【摘要】 目的 :考察黄芪提取物对犬心力衰竭模型的作用。方法 :分别用戊巴比妥钠和维拉帕米造成犬急性心力衰竭模型 ,以 0 .8、0 . 4和 0 . 2mg/kg黄芪提取物注射液为受试药 ,观察模型犬于造模前后及给药后 5、10、30、6 0和 90分钟时 ,血流动力学指标动脉收缩压 (SAP)、心率 (HR)、心输出量 (CO)、左心室内收缩压 (LVSP)和左心室内压最大上升速率 (LV +dp/dtmax)的变化 ,其中以LV +dp/dtmax下降 5 0 %~ 75 %作为心力衰竭指标。结果 :在戊巴比妥钠致心力衰竭模型实验中 ,与模型对照组相比 ,黄芪提取物注射液高、中剂量组在给药后各时间点的SAP明显增加 (P <0 . 0 5 ) ,高、低剂量组在给药后 30、6 0和 90分钟时的HR明显增加 (P <0 . 0 5或P <0 . 0 1) ,高剂量组在给药后各时间点的CO明显增加 (P <0 .0 5 ) ,高剂量组在给药后 5、10、30和 90分钟时的LVSP明显增加 (P <0 . 0 5或P <0 . 0 1) ,高剂量组在给药后 5、10、6 0和 90分钟时的LV +dp/dtmax分别增加了 14 . 4 %、17 .79%、2 3 .2 4 %和 2 8. 97%(P <0 . 0 5 ) ;在维拉帕米致心力衰竭模型实验中 ,与模型对照组相比 ,高、低剂量组在给药后 30、6 0和 90分钟时的SAP明显增加 (P <0 .0 5 ) ,高剂量组在给药后 10分钟的HR明显增加 (P <0. 0 5 ) ,高剂量组

【Abstract】 Objective: To investigate the effect of Astragalus extract (AE) on acute heart failure in dogs. Methods: The dog models of acute heart failure were prepared with pentobarbital sodium and verapamil respectively. Their values of SAP, HR, CO, LVSP and LV+dp/dt max were measured respectively before and after modeling, and at 5, 10, 30, 60, 90 min after administration of 0.8, 0.4 and 0.2 mg/kg AE. The decline in LV+dp/dt max by 50%-75% was taken as the mark of heart failure. Results: In the experiment in dog models of heart failure induced by pentobarbital sodium,compared with model control group, 0.8 and 0.4 mg/kg AE increased significantly the value of SAP at each time point after administration(P<0.05); 0.8 and 0.2 mg/kg AE raised significantly the value of HR at 30, 60 and 90 min after administration(P<0.05 or P<0.01); 0.8 mg/kg AE improved significantly the value of CO at each time point after administration(P<0.05); 0.8 mg/kg AE hightened significantly the value of LVSP at 5,10,30 and 90 min after administration(P<0.05 or P<0.01); 0.8 mg/kg AE enhanced significantly the value of LV+dp/dt max by 14.4%, 17.79%, 23.24% and 28.97% at 5, 10, 60 and 90 min after administration(P<0.05). In the experiment in dog models of heart failure induced by verapamil,compared with model control group, 0.8 and 0.2 mg/kg AE increased significantly the value of SAP at 30, 60 and 90 min after administration(P<0.05); 0.8 mg/kg AE raised significantly the value of HR at 10 min after administration(P<0.05); 0.8 mg/kg AE hightened significantly the value of LVSP and LV+dp/dt max, the latter by 7.17%, 20.53%, 23.88% and 30.39%, at 10, 30, 60 and 90 min after administration(P<0.05或 P<0.01); but every group has no effect on the value of CO at each time point after administration(P>0.05). Conclusion:AE has antagonistic effect on heart failure induced by pentobarbital sodium and verapamil.
