鲨鱼肝再生因子对肝细胞再生的作用
Studies on the Mechanism of Shark Hepatic Regenerative Factor on Rat Liver Regeneration
【摘要】 通过给切除部分肝脏大鼠注射鲨鱼肝再生因子,观察其对肝脏细胞再生的促进作用,为临床用药提供依据。按Higgins和Anderson方法对大鼠行肝切除术,分别尾静脉注射鲨鱼肝再生因子(SHRF)12,6,3 mg/kg,促肝细胞生长素(HGF 6 mg/kg),生理盐水(0.3 ml/100 g)术后10 d取肝脏称重及记录。并对各时相点大鼠取血和肝细胞培养。比较各组大鼠血清甲胎蛋白(AFP)和肝细胞中一氧化氮(NO)含量的变化。结果给药组与模型组肝脏重量比较有极显著差异,给药组和阳性药组血清中AFP及肝细胞中NO含量与模型组相比均呈升高趋势,因此SHRF在短期内对大鼠肝脏部分切除术后再生有明显的促进作用。
【Abstract】 The SHRF in rats of partical hepatectomy was investigated to explore the mechanism of liver regeneration compared with positive group.It may be used as a reference for clinical application.SD rats were divided into six groups randomly: model group,sham operation group,positive group(HGF),high dose group,middle dose group,and low dose group(SHRF),with ten rats in each group.And we injected NaCl in model and sham operation group and injected SHRF,HGF in the other four groups at 24h before operation,and at 24h and 48h after operation.The other five groups need to adopt the methods of hepatectomy of Higgins and Anderson except sham operation group in which opening abdominal and then closing it are needed.Ten days after the opteration we weighted the liver and made a record.Wister rats were divided into six groups randomly.Each group was divided into eight phases randomly again,which was 0h,12h,24h,36h,48h,72h and 1 week after partial hepatectomy.At the same time,we obtained the blood in the eyes and used the technology of cells culture to determine the level of NO in hepatic cell and the level of AFP in blood at respective time.The level of AFP showed the trend of increase after operation,and continued for 1 week.SHRF and positive drug could enhance the synthesis of AFP after partial hepatectom.The level of NO began to increase at 1h after operation,obtained a peak at 6h,a minimum at 12h,and a maximum(again) from 30h to 37h.SHRF can improve the level of AFP,NO in 1 week.And the content of NO had the relativity with the time.It can stimulate the liver regeneration.
