离子抑制-反相高效液相快速测定池杉、落羽杉根系中有机酸
Rapid Determination of Rudimental Organic Acids in Root of Taxodium ascendens and Taxodium Distichum by Ion-suppression RP-HPLC
【摘要】 目的:建立用离子抑制-反相高效液相色谱快速测定池杉、落羽杉根系中 L-苹果酸、莽草酸含量。方法:采用 Agilent1100 DAD 检测器,Hypersll ODS2 5 μm(4.6mm×150mm)柱,HClO4(pH=2.5)溶液为流动相,柱温30℃时于214 nm 波长处以外标法测定。结果:L-苹果酸、莽草酸分别在7.16×10-6mol·L-12.24×10-2mol·L-1和1.27×10-7mol·L-1~2.06×10-4mol·L-1范围内,浓度与峰面积有良好的线性关系(r=1.0000和r=0.9999)。检测限分别为2.42×10-8mol·L-1和1.16×10-9mol·L-1(S/N=3)。结论:该法简便、快速、有效、重现性好,首次用于实际池杉、落羽杉根系中 L-苹果酸、莽草酸的测定,取得了很好的结果。
【Abstract】 Objective:A ion-suppression reversed-phase HPLC method was established for the determination of rudimental L-malic acid and Shikimic acid in Taxodium aseendems and T axodium distiehum.Methods:They can be separated on Agilent 1100 DAD and Hypersil ODS2 5μm,(4.6 mm×150 mm)column by using perehloric acid (pH=2.25)as mobile phase with UV detector at a wavelength of 214 nm.Results:The standard curve of L-malic acid and shikimic acid was linear in the concentration range 7.16×10-6mol·L-1-2.24×10-2mol·L-1 and 1.27×10-7 mol·L-1-2.06×10-4mol·L-1and the correlation coefficient was 1.0000 and 0.9999.The detec- tion limit was 2.42×10-8 mol·L-1 and 1.16×10-9 mol·L-1(S/N=3).Conclusions:The method has been suc- cessfully applied to the determination of trace rudimental L-malic acid and Shikimic acid in the root of Taxodium ascendens and Taxodium distiehum,which was proved to be accurate,simple effective and feasible.
