【摘要】 目的探讨转化生长因子β1(TGFβ1)诱导人视网膜色素上皮(RPE)细胞凋亡过程中Survivin基因的表达及意义。方法用免疫组织化学染色法检测人RPE细胞Survivin蛋白的表达;用AnnexinVFITC标记法测定TGFβ1作用下的人RPE细胞凋亡发生率;用RTPCR方法检测不同质量浓度TGFβ1作用下人RPE细胞Survivin基因的表达。结果免疫化学染色法显示:人RPE细胞表达Survivin基因;AnnexinVFITC标记法检测结果显示:人RPE细胞的凋亡率随着TGFβ1质量浓度的升高而增高;RTPCR结果显示:随着TGFβ1质量浓度的升高,Survivin基因的表达逐渐降低。结论Survivin基因在人RPE细胞的凋亡过程中起着非常重要的作用。更多还原

【Abstract】 ObjectiveTo investigate the expression of Survivin gene in TGF-β_ 1 -mediated apoptosis of hRPE cells.MethodsImmunochemistry staining with SABC was used to detect the expression of Survivin mRNA in hRPE cells,and apoptosis rate of RPE cells induced by TGF-β_ 1 was measured using Annexin V-FITC labeling method.The expression of Survivin gene in hRPE cells was detected by RT-PCR technique.ResultsExpression of Survivin gene was detected in RPE cells.The apoptosis rate of hRPE cells was positive correlated with TGF-β_ 1 concentration,and the expression of Survivin mRNA was markedly reduced with increase of TGF-β_ 1 level.ConclusionSurvivin gene plays an important role during the apoptosis of hREP cells.更多还原
