视网膜脱离时神经生长因子对Mǖller细胞中间丝蛋白表达的影响
Effect of nerve growth factor on the expression of intermediate filament proteins in Müller cells in experimental retinal detachment
【摘要】 目的研究实验性视网膜脱离(RD)时外源性神经生长因子(NGF)对M櫣ller细胞功能的影响作用。方法29只SD大鼠建立RD模型,并分为NGF实验组、空白、单纯RD、正常对照共4组。在建立RD模型后观察1.5、3、6h,1/2、1、2、4、8、16、32d。NGF实验组在建立模型后每4d在玻璃体腔内注射NGF5μg/5μL/次,空白对照组注射空白载体作为对照。采用免疫组织化学方法对M櫣ller细胞的GFAP和vimentin进行标记。非参数统计方法分析。结果单纯RD组显示随脱离时间延长,二者在M櫣ller细胞的表达强度逐渐增强,分布范围变广。NGF实验组显示M櫣ller细胞内的二者表达强度低于对照组,差异有显著性(P<0.05)。结论外源性NGF能抑制RD后M櫣ller细胞过度表达GFAP和vimentin,防止M櫣ller细胞过度反应。
【Abstract】 ObjectiveTo study the effect of nerve growth factor (NGF) on Müller cells in experimental retinal detachment (RD).MethodsTwenty-nine Sprague-Dawely rats were used for an animal model of RD induced by infused sodium hyaluronate under the neurosensory retina.Rats were divided into a NGF group,a blank control group,a RD group and a normal control group.5μg/5μL of NGF was intravitreously injected in NGF group,and 5μL of PBS was used in the blank control group after a time course of 4-days of RD.Animals were sacrificed and the eyeballs were investigated at 1.5,3,6 hours and 0.5,1,2,4,8,16,32 days,respectively for the identification of changes of Müller’s intermediate filament proteins by immunohistochemistry.A non-parameters method was used for the statistical analysis.ResultsLow positive staining intensity of glial fibrillary acidic protein (GFAP) was seen and vimentin were expressed in endfoot region of normal Müller cells.In RD control group,the staining intensity was gradually increased in the detached retina,and was significantly higher than that in normal group even after retina reattached.In NGF groups,staining intensity were significantly lower than that in blank group (P<0.05).ConclusionAfter retina detached,exogenous NGF can suppress expressions of GFAP and vimentin in Müller cells,implying that NGF can inhibit over reaction of Müller cells.
