阿司匹林和肿瘤坏死因子α对脂肪细胞脂肪源性细胞因子表达的影响
Effects of aspirin and tumor necrosis factor-α on the gene expression of adipocytokines in adipocytes
【摘要】 目的观察阿司匹林和肿瘤坏死因子α(TNF α)对脂肪细胞脂肪源性细胞因子表达的影响。方法5 mmol/L阿司匹林和/或5 ng/ml TNF α处理3T3 L1脂肪细胞48 h后抽提总RNA,用半定量逆转录聚合酶链反应(RT PCR)和Northern印迹检测脂肪细胞脂源性细胞因子的表达。结果TNF α抑制了抵抗素、脂联素基因的表达,促进了白细胞介素6(IL 6)、纤溶酶原激活物抑制剂1(PAI 1)基因的表达;阿司匹林对上述基因的表达并无影响,但部分拮抗了TNF α对抵抗素、脂联素基因的抑制。结论TNF α抑制了抵抗素、脂联素基因的表达,促进了IL 6、PAI 1 基因的表达,可能引起机体的胰岛素抵抗(IR)和相关的心血管并发症;阿司匹林部分拮抗了TNF α对抵抗素、脂联素基因的抑制可能与其改善IR及相关心血管并发症有关。
【Abstract】 Objective To observe the effects of aspirin and tumor necrosis factor-α(TNF-α) on the gene expression of adipocytokines in adipocytes. Methods Total RNA of 3T3-L1 adipocytes incubated with 5mmol/L aspirin and/or 5ng/ml TNF-α for 48 hours was isolated by TRIZOL method. Semi-quantitative RT-PCR and Northern blot were performed to quantitate the mRNA expressions of adipocytokines. Results mRNA levels of resistin and adiponectin were decreased and the levels of IL-6 and plasminogen activator inhibitor-1(PAI-1) were increased in 3T3-L1 adipocytes incubated with TNF-α for 48 hours. Aspirin did not influence on resistin,adiponectin,interleukin-6(IL-6) and PAI-1 gene expressions,while alleviated the inhibiting effect of TNF-α on resistin and adiponectin expression. Conclusion TNF-α inhibits resistin and adiponectin gene expressions, but increases IL-6 and PAI-1 gene expressions,which may contribute to IR and cardiovascular complications. Aspirin alleviates the inhibiting effects of TNF-α on resistin and adiponectin expression, which may be related to improving insulin sensitivity and cardiovascular complications.
