LPS对人脐静脉内皮细胞增殖和ESM-1表达的影响
LPS effects on the growth of human umbilical vein endothelial cells and expression of ESM-1
【摘要】 目的 探讨LPS对人脐静脉内皮细胞(ECV304)的生长和内皮细胞特异性分子 1 (endothelialcell specificmole cule 1,ESM 1)表达的影响。方法 在体外,用 1ml/L和 5ml/L的LPS在不同时间作用于ECV304,观察细胞形态的变化,用MTT法和免疫细胞化学方法对细胞增殖和ESM 1、ERK、p ERK在细胞中的表达。结果 LPS作用于ECV30424h后,细胞形态发生变化,梭形明显,排列紊乱。短时间内(0 5、1h)LPS对ECV304有增殖作用。LPS可快速促进ESM 1的表达,在 0 5、1h时表达量较高。LPS作用后,ERK的表达无明显变化,但在一段时间 ( 0 5 ~4h)内ERK的磷酸化明显增加。结论 LPS对ECV304中ESM 1的表达有促进作用。推测ESM 1可能作为一种细胞因子与LPS在炎症反应中发挥着作用。
【Abstract】 Objective To study the effects of LPS on the growth of human umbilical vein endothelial cells and expression of endothelial cell specific molecule 1(ESM 1). Methods The endothelial cells were cultured by 1 ml/L and 5 ml/L LPS at different time intervals. The morphology of endothelial cells was observed and the cell proliferation was measured by MTT method. The expressions of ESM 1, ERK and p ERK were detected by immuocytochemistry. Results The morphology of endothelial cells was altered to become long cord like in shape and array in disorder by LPS after 24 h. The cell proliferation was stimulated by LPS during 1 h. The expression of ESM 1 was quickly enhanced at 0 5 h and significantly at 1 h. There was no change of expression of ERK, but ERK phosphorylation was remarkable increased at 0 5 h to 4 h. Conclusion LPS can stimulate expression of ESM 1 in endothelial cells. ESM 1 may play an important role in inflammatory reaction as a cell factor with LPS concomitantly.
