补充谷氨酰胺对运动大鼠骨骼肌desmin表达的影响
The Effect of Glutamine Supplementation on Regenerated Skeletal Muscle Fibers of Exercised Rats
【摘要】 目的 :观察补充谷氨酰胺对运动大鼠骨骼肌结蛋白 (desmin)表达的影响 ,探讨补充谷氨酰胺提高运动能力的作用机制 ,以及补充谷氨酰胺的适合时间。方法 :32只成年雄性SD大鼠随机分为安静对照组 (6只 )、运动 2天补充安慰剂组 (2s组 ,6只 )、运动 5天补充安慰剂组 (5s组 ,7只 )、运动 2天补充谷氨酰胺组 (2g组 ,6只 )、运动 5天补充谷氨酰胺组 (5g组 ,7只 )。运动组每天进行90分钟的下坡跑 ,坡度 - 16° ,速度 16~ 18米 /秒。运动后通过灌胃方式补充谷氨酰胺 ,平均剂量为1克 /千克体重·每天 ,分别在运动后即刻和第 1次补充后的 6~ 8小时分 2次补充。通过免疫组织化学方法对大鼠比目鱼肌中desmin进行染色 ,以鉴别卫星细胞的激活和新的肌纤维的生长。结果 :谷氨酰胺能促进再生肌纤维中desmin的合成 ,且主要发生在运动 2天后。连续运动 2天后 ,补充谷氨酰胺组的desmin阳性肌纤维横截面积较补充安慰剂组显著增加。结论 :补充谷氨酰胺有助于促进运动性肌肉微损伤后的恢复 ,补充的合适时间在连续运动的 5天内。
【Abstract】 Objective To investigate the effect of glutamine su pplementation on e xpression of desmin in skeletal muscle in exercised rats, and to explore the mec hanism of improved performance through glutamine supplementation and the appropr iate timing for glutamine supplementation. Methods Thirty-two adult male Sprague-Dawley rats were divided randomly into sedentary control grou p, 2-day-exercise-placebo group (2s group), 2-day-exercise-glutamine group (2g g roup), 5-day-exercise-placebo group (5s group) and 5-day-exercise-glutamine grou p (5g group). The animals in exercised group were running at a speed of 16~18m/ min on a treadmill with decline slope of 16° for 90 minutes.The average dosage of supplemental glutamine was 1g/kg body weight/day. The first supplementation was given immediately after the exercise, and the second was given 6~8 hours followed the first one. In order to identify the activation of satellite cells an d the development of regenerated fibers, sections of rat soleus was stained for desmin by immunohistochemical technique. Results Glutamine could facilitate the expression of desmin in regenerated muscle fiber, which occ ured mainly within 2 days post exercise. After 2 days successive exercise, th e cross section area of desmin-positive fiber of soleus in glutamine-supplemente d rats were greater than that in placebo-supplemented rats. Conclusion Glutamine supplementation accelerates the recovery process of exercise-induc ed muscle damage. The appropriate timming of glutamine supplementation is within 5 days during continuous exercise.
