丹参注射液限制犬心肌梗死范围及其机制研究

【摘要】 目的探讨丹参注射液对心肌梗死(M I)犬心的保护作用及其部分机制。方法10只犬随机分成丹参组(D组)和对照组(C组),建立M I模型,分别给予丹参注射液和生理盐水,共21 d,冠脉结扎前、后行冠脉造影,术前及术后d22查8项血液流变学指标,测量M I面积及心肌血管面密度。结果①D组M I面积明显小于C组。②冠脉结扎后2h和22 d D组侧枝血管明显多于C组。③在M I急性期,血小板聚集性和血粘度较M I前明显增高,而丹参注射液能阻止上述改变。④D组缺血区心肌毛细血管和供血血管面密度较C组明显增加。结论丹参注射液能促进犬冠脉侧枝循环的开放和形成,抑制M I后血小板聚集和降低血粘度,改善微循环,限制M I范围。

【Abstract】 Aim To evaluate the protective effect of salvia miltiorrhiza(SM) on ischemic myocardium in dogs with acute myocardial infarction(AMI) and to reveal its possible mechanism. Methods Ten dogs were randomly divided into SM(D)group and control(C)group. AMI model was built in all dogs, SM or saline was administrated for 21 days. Coronary angiography were performed before and after ligation of coronary artery. 8 hemorheological parameters were examined before and 22 d after the operation. The infarct zone and vessel density of myocardium were assessed.Results ①The infarct area in group D was smaller than that in group C. ②Angiography at hour 2 and on day 22 after ligation of coronary artery revealed significant augmentation of collateral vessels in group D versus group C. ③The platelet aggregation and the blood viscosity increased during AMI when compared with control phase, and the increased indicators during AMI were inhibited when SM was given. ④Capillaries and distribution vessel density in ischemic site on day 22 appeared statistically significant augmentation in group D as compared with the control group. Conclusion SM might improve the opening and formation of coronary collateral circulation, and inhibit the increase of platelet aggregation and blood viscosity during AMI, and then improve microcirculation and restrict myocardial infarct area.
