植物雌激素α-ZAL对oxLDL诱导的人内皮细胞分泌NO和ET-1的影响及其机制研究
Effect and mechanism of phytoestrogen α-Zearalanol on NO and ET-1 secretion induced by oxLDL in human umbilical vein endothelial cells
【摘要】 目的探讨植物雌激素α-ZAL影响oxLDL诱导的人脐静脉内皮细胞分泌NO、ET-1的机制。方法采用RTPCR方法对eNOS、iNOS、ET-1mRNA的表达进行分析;采用WesternBlotting方法对eNOS、iNOS蛋白表达进行分析;同时测定内皮细胞上清液中NO、ET-1分泌的变化。结果植物雌激素α-ZAL增加oxLDL诱导内皮细胞eNOSmRNA、蛋白的表达和NO的分泌,降低ET-1mRNA的表达和蛋白的分泌。结论植物雌激素α-ZAL通过上调oxLDL诱导内皮细胞eNOSmRNA、蛋白的表达和NO的分泌,下调ET1mRNA的表达和蛋白的分泌起着保护血管内皮的作用。
【Abstract】 Aim To evaluate the effect of a novel phytoestrogen α-Zearalanol on oxLDL-induced effect on NO and ET-1 production in human umbilical vein endothelial cells.Methods Protein and mRNA expression of NOS and ET-1 were measured by Western blot and RT-PCR, levels of NO and ET-1 were measured by spectrophotometry and enzymatic immunoassay.Results Our results indicated that oxLDL significantly reduced NO release and enhanced ET-1 production associated with reducing eNOS protein/mRNA expression and enhancing ET-1 mRNA expression. α-Zearalanol could attenuate or abolish these oxLDL-induced alterations.Conclusion Phytoestrogen α-Zearalanol may effectively antagonize oxLDL-induced decrease in NO and increase in ET-1, which may be protective for endothelial function.
