双黄连对人巨细胞病毒感染小鼠肝组织病毒DNA载量影响
The effect of Shuanghuanglian on the load of hepatic HCMV DNA in mice infected with HCMV
【摘要】 目的研究双黄连(Shuanghuangliang,SHL)粉针剂对人巨细胞病毒(humancytomegalovirus,HCMV)感染的小鼠肝组织病毒DNA载量的影响。方法以HCMV感染小鼠建立小鼠肝炎模型后,用不同剂量双黄连腹腔注射进行治疗。用药10d后,颈椎脱臼处死动物取肝脏,用荧光定量PCR的方法检测小鼠肝组织HCMVDNA载量;HE染色法对肝脏组织作病理检查;眼眶静脉丛采血用于ALT、AST酶活性的测定。结果双黄连大、中剂量治疗组肝组织中HCMVDNA载量平均值分别为8.3261copies/mg、9.1476copies/mg,较感染模型组(载量平均值为3875.4760copies/mg)降低(P<0.01),与更昔洛韦(ganciclovir,DHPG)治疗组(载量平均值为9.3280copies/mg)差异无显著性(P>0.05),双黄连小剂量组(载量平均值为3812.8980copies/mg)与感染模型组差异无显著性(P>0.05);双黄连大、中剂量治疗组肝脏ALT、AST水平较模型组降低,病理损伤明显减轻;小剂量治疗组与感染模型组无差异;双黄连大、中剂量组和更昔洛韦组差异无显著性。结论双黄连能明显降低HCMV感染小鼠肝组织中病毒DNA载量,能减轻HCMV性肝炎模型鼠肝功能损害,改善肝脏病理变化。
【Abstract】 Aim To investigate the effect of Shuanghuanglian on the the load of hepatic HCMV DNA in mice infected with HCMV.Method The model of mice hepatitis infected with HCMV was prepared. 10 days after treated with Shuanghuanlian,the hepatic HCMV DNA was detected qualitatively by polymerase chain reaction;the level of alanine aminotransferase (ALT) and aspartate aminotransferase(AST) in serum were assayed by spectrophotometer;and hepatic pathology was observed with HE staining.Results HCMV DNA load in infected model group was 3875.4760 copies/mg,while in high and middling dosage shuanghuanglian therapy group it was 8.3261 copies/mg、9.1476 copies/mg respectively;It decreased significantly compared with the former(P<0.01).No significant differences were explored between ganciclovir therapy group and high and middling dosage groups (P>0.05);low dosage of shuanghuanglian(HCMV DNA load was 3812.8980 copies/mg) had no notable difference compared with infected model group;serum transaminase (ALT,AST) levels significantly elevated in infected model group; serum transaminase level decreased sharply and liver histological pathology was lighten after treated with high and middling dosage of Shuanghuanglian(P<0.05);Conclusion Shuanghuanlian can reduce the load of the hepatic HCMV DNA and alleviate liver lesion and lighten liver histological pathology change in HCMV hepatitis model,its effect is similar to that of ganciclovir.
