【摘要】 目的对Ad-PDZ1-GFP穿梭重组体与骨架载体进行原核重组和真核包装。方法将Ad-PDZ1-GFP穿梭重组体与骨架载体以电穿孔法共转染至原核包装细胞BJ5183;再将原核重组体用脂质体转入真核包装细胞293H进行包装。结果①电泳鉴定得到原核重组体;②得到完整的具感染能力的病毒颗粒;③病毒颗粒能表达标签蛋白。结论获得了具有感染能力的病毒颗粒。更多还原

【Abstract】 Objective To construct recombinant pAdTrack-CMV-PDZ1.?Methods Using the total RNA extracted from fresh rat hippocampus as template,the cDNA of PDZ1 was amplified by RT-PCR;the cDNA obtained was subsequently cloned into the shuttle vector pAdTrack-CMV(CLONTECH),and then,the sequence of the product was determined.?Results ①Total RNA was obtained.②The cDNA of PDZ1 was amplified by RT-PCR.③The recombinant was found containing two bands of pAdTrack-CMV/PDZ1 by restriction enzyme digestion.④The sequence of cloned PDZ1 was identical with that reported in GeneBank.?Conclusion Recombinant of pAdTrack-CMV-PDZ1 is obtained.更多还原
