对抑郁模型小鼠强迫游泳实验方法的探讨
The conditions of forced swimming test in mice
【摘要】 目的研究小鼠强迫游泳实验方法的条件,探讨在不同条件下小鼠对这种抑郁模型的敏感性是否有差别。方法采用ICR雄性小鼠,分为给药组(盐酸丙咪嗪)和对照组,通过强迫游泳实验,观察预先训练、体重和水深对不动时间的影响。结果经过游泳训练的小鼠给药组的不动时间比对照组短,未经游泳训练的小鼠2组间差异无显著性。体重20~24g的小鼠中,给药组与对照组间差异有显著性;体重24~28g的小鼠中,2组间差异无显著性。水深为10cm时,大剂量(10mg·kg-1)、小剂量(5mg·kg-1)给药组与对照组间差异均有显著性;水深为6cm和15cm时,仅大剂量(10mg·kg-1)给药组与对照组间差异有显著性。结论小鼠强迫游泳实验以选择体重20~24g的ICR雄性小鼠、经过训练、水深为10cm为宜。
【Abstract】 Objective To investigate the effects of the conditions of forced swimming test on the behavioral despair in model mice. Methods Male ICR mice were divided into drug groups (imipramine hydrochloride) and control groups (distilled water) to undergo forced swimming test. The conditions to be examined in the test included experience of training, body weight and the depth of water in the swimming jar. Results The immobility time (ImT) was prolonged significantly after training. Administration of immipramine 10 mg·kg~-1 shortened ImT in trained mice(P<0.05), but not in the untrained mice. The ImT became shortened after the drug administration in the 20~24 g trained mice(P<0.01), not in the older 24~28 g ones(P>0.05). Ten cm was found to be the choice depth of water, where the test was sensitive even if the drug was given smaller dose (5 mg·kg~-1 ,P<0.05), while in case of 6 cm and 15 cm depths, conclusive results were only obtained in 10 mg·kg~-1 group(P<0.05). Conclusion Trained male mice, body weight 20 g to 24 g and the depth of water at 10 cm, are the suitable conditions for forced swimming test.
