徐州地区汉族青年淋巴细胞对糖皮质激素反应的差异
The differences in lymphocyte steroid sensitivity among healthy youths of Han nationality in Xuzhou
【摘要】 目的通过检测地塞米松(Dex)对体外培养的120名徐州地区健康的汉族青年外周血淋巴细胞(PBLs)活化及分泌的影响,探讨该群体对糖皮质激素(GC)反应敏感性(LSS)的差异。方法以PBLs为研究标的细胞,用植物血凝素(PHA)及不同浓度Dex(10-8~10-6mol/L)处理,培养72h后用流式细胞仪检测CD3、CD25表达率,以酶联免疫吸附(ELISA)法检测白细胞介素-2(IL-2)水平,3个月后重复实验。分别计算GC对PBLs活化和分泌的最大抑制率(Imax1和Imax2)。结果根据Imax1,分别有28人和87人表现为GC抵抗和GC敏感;根据Imax2,分别有25人和88人表现为GC抵抗和GC敏感;两项指标结果一致的有103人,其中23人均表现为GC抵抗,80人均表现为GC敏感。不同浓度Dex均可显著抑制GC敏感者PBLs活化和分泌(P<0.01),并呈现剂量依赖关系;GC抵抗者不同浓度Dex组之间PBLs活化和分泌均无显著性差异;同一个体2次实验Imax的差异显著小于同一次实验中不同个体Imax的差异(P<0.01)。结论Imax是反映LSS的一项非常稳定的参数,可以估计个体PBLs对GC的反应。徐州地区汉族人的LSS存在差异性,结合活化和分泌两项指标,筛选出的GC抵抗者高达20.9%,说明正常人群中存在一定比率的对GC抵抗的个体,推测他们的遗传背景可能与GC敏感者不同。
【Abstract】 Objective To investigate the variation in lymphocyte steroid sensitivity (LSS) on activation and secretion in vitro among healthy human volunteers from Xuzhou,China.?Methods Peripheral blood lymphocytes (PBLs) from 120 young volanteers of Han nationality, were used as the target cells. They were treated with phytohemagglutinin (PHA) and dexamethasone (Dex) at concentrations of 10~-8 to 10~-6 mol/L.After 72 h of cultivation, the interleukin-2 (IL-2) level was determined by ELISA,the expression of CD3 and CD25 by flow cytometry. All of the assessments were repeated three months later. Maximal percentage inhibition (I_~max ) was defined as the maximal percentage inhibition either of secretion or activation achieved at different concentrations of Dex. Taking 50% as the set point,the individuals who failed to achieve more than 50% inhibition were considered as steroid-resistant (SR), and those whose inhibition exceeded 50% as steroid-sensitive (SS).?Results Analysed results showed there were 80 SS and 23 SR.Dex of different concentrations inhibited the activation and secretion in a dose-dependent manner (P<0.05), while in the SR group,no such findings were noticed.The I_~max values,both of activation and secretion,might deviate widely between individuals in the same SS group, but remained stable in a given individuals over time.?Conclusion I_~max is a stable parameter responsible for LSS in vitro.The fault of LSS may be widely present in healthy people, as up to 20.9% of healthy Han youth in Xuzhou were found to be SR.It is believed that hereditary background may play an important role in variation of LSS.
