门冬氨酸钾镁对肝细胞代谢的影响

【摘要】 目的:探讨门冬氨酸钾镁溶液对人肝细胞代谢的影响及其相关机制。方法:对培养的人肝细胞L 02株分为 3组,分别于培养基中加入门冬氨酸钾镁复盐溶液(研究组),硫酸镁与氯化钾的混合液(对照组 )及空白对照 (空白组 )。测定肝细胞内K+,Mg2+的浓度;检测肝细胞胞膜Na+ K+ ATP酶活性;测定ATP含量及线粒体琥珀酸脱氢酶(SDH)、细胞色素氧化酶 (CCO)的活力。加入适量铵盐后测定各组NH4+和尿素的浓度以了解各组肝细胞对氨的清除能力。结果:与空白组和对照组相比,研究组的K+,Mg2+进入细胞内的量明显增多(P<0.05或P<0. 01 );其胞膜的Na+ K+ ATP酶活性升高(P<0. 01);氨清除加快,尿素生成增多(P<0.05或P<0.01);ATP浓度升高 (P<0. 01),与能量代谢密切相关的线粒体CCO活性和SDH活性增高,能量代谢能力明显增强 (P<0. 01)。结论:门冬氨酸钾镁溶液能显著增强人肝细胞的代谢能力。

【Abstract】 AIM: To investigate the effects of potassium-magnesium aspartate injection on hepatic cells metabolism and its correlative mechanism. METHODS: The cultured human hepatic cells were divided into three groups, the potassium-magnesium aspartate injection group (research), mixture of MgSO 4 and KCl solution group (control), and buffer solution group (blank). The K +, Mg 2+ concentrations in hepatic cells, the activity of Na +-K +-ATP ase in hepatic cell-membrane, the ATP level, succinat e dehydrogenase (SDH) and cytochrome c oxidase (CCO) activities in mitochondria were assayed. After adding appropriate amount of NH + 4 6 h into the culture, the NH + 4 and urea concentrations in supernatant of cultured hepatic cells were assayed for measuring the NH + 4 clearance and urea production by hepatic cells. RESULTS: Comparing with the blank group, control group and the research group significantly promoted the hepatic cells metabolism, with the elevations of entry amount of K +, Mg 2+ into cells (P<0.05 or P<0.01), the activities of Na +-K +-ATP ase, SDH, CCO and the ATP contents (P<0.01). The NH + 4 clearance and urea production were also enhanced by potassium-magnesium aspartate (P<0.05 or P<0.01). CONCLUSION: Potassium-magnesium aspartate solution significantly enchances the metabolic potency of human liver cells.
