【Abstract】 AIM: To explore the effects of different doses clozapine on the glucose metabolism in male schizophrenics. METHODS: Eighty-seven male schizophrenics were randomly divided into three doses groups: the high dose group took clozapine over 300 mg·d -1; the middle one 150-300 mg·d -1, and the low one less than 150 mg·d -1. Before and after 6 wk treatment, all the patients were given oral glucose tolerance test (OGTT), with blood glucose measuring at 0,1,2,and 3 h. The body mass index (BMI) was also calculated. The data were analysed with t,χ2,q and ANOVA test. RESULTS: After 6 wk treatment, the blood glucose contents of the high dose group at the time-points of 1 and 2 h were higher than those of the baseline ones, respectively(P<0.01), while the other two groups showed no changes. Among three groups, the significant difference were only found between the high dose and the low dose groups (P< 0.05). The rate of impaired glucose tolerance (IGT) at the time-point of 1 h was higher in high dose group than in low dose group after treatment (P<0.05).Weight gains of the high dose groups were obviously increased after treatment(P<0.01). There was no significant relationship between the weight gains and the IGT in three groups before and after treatment. CONCLOSION: The high dose clozapine has stronger effects on the glucose metabolism and weights of male schizophrenics.
