姜黄素影响Caspase8和Caspase9诱导淋巴瘤Raji细胞凋亡的研究
Effect of Curcumin on Caspase 8- and Caspase 9 -induced Apoptosis of Lymphoma Raji Cell
【摘要】 姜黄素(curcumin)是中药姜黄的主要成分,能选择性地抑制肿瘤细胞增殖并诱导其凋亡,本研究旨在探讨姜黄素抗癌作用的分子机制。选择淋巴瘤细胞系Raji为研究对象,用正常健康人静脉血作为对照,应用淋巴细胞分层法分离获得外周血单个核细胞(PBMNC)并进行培养。用四甲基偶氮唑蓝(MTT)比色法检测姜黄素不同浓度、不同时间作用于Raji细胞、PBMNC后细胞的抑制率,并进行比较。用Westernblot检测25μmol/L(IC50)姜黄素作用24小时Raji细胞胱冬蛋白酶Caspase8和9的表达。结果表明:姜黄素可呈时间及剂量依赖性抑制Raji细胞增殖,可显著促进Raji细胞凋亡(P<0.01),且呈浓度依赖性及时间选择性;一定浓度范围内的姜黄素对PBMNC无显著抑制作用,提示姜黄素抑制增殖具有选择性作用。Raji细胞胱冬蛋白酶8和9的表达明显增高(P<0.05)。结论:胱冬蛋白酶8和9在Raji细胞增殖和凋亡中起重要的作用,姜黄素在不同浓度、不同时间条件下抑制Raji细胞增殖并促进其凋亡。
【Abstract】 Curcumin is a crucial component of curcuma. Recently more attention has been paid to the effect of curcumin on specific proliferative inhibition and inducing apoptosis of tumor cells. This study was aimed to investigate the anticancer activities of curcnmin and its molecular mechanism. Raji cells (lymphoma cell line) were selected as studying targets, peripheral blood mononuclear cells (PBMNC) obtained from healthy donors were separated by Ficoll solution and suspended in RMPI 1640. The inhibition rates of Raji cells and PBMNC after treatment with curcumin at various concentrations and different times were determined by MTT method and were compared. The expressions of caspase 8 and caspase 9 in Raji cells after treatment with curcumin at 25 μmol/L (IC50) and for 24 hours were detected by Western blot. The results showed that curcumin could inhibit proliferation of Raji cells in dose-and time-dependent manner. Cucurmin could remarkablely enhance the Raji cell apoptosis at 25 μmol/L and 24 hours (P<0.01), and its effect was dose-dependent and time-selective. Curcumin had no remarkable effect on PBMNC at certain concertrations,which demonstrated that curcumin could selectively inhibit tumor cell proliferation. It is concluded that the expression of caspase 8 and caspase 9 plays an important role in the proliferation and apoptosis of Raji cells, so that curcunin showed inhibitive effect on Raji cells at various concentrations.
