反相高效液相色谱法测定茯苓中茯苓酸的含量

【摘要】 目的建立茯苓中茯苓酸的含量测定方法。方法采用反相高效液相色谱法(RP-HPLC),HypersilODSC18色谱柱(250mm×4.6mm,5μm),柱温30℃,乙腈-0.5%磷酸(70∶30)为流动相,流速0.8mL/min,检测波长242nm。结果茯苓酸在0.362~9.050μg范围内与其色谱峰面积呈良好线性关系,r=0.9999,平均回收率99.54%,RSD=1.01%。3个产地茯苓中茯苓酸的含量依次为:湖北>云南>安徽。结论该方法简便、专属、稳定,可用于茯苓药材的质量评价。

【Abstract】 Objective To establish a method for determining content of Pachymic acid in Poria.Methods RP-HPLC method was used for determination of Pachymic acid in Poria from Anhui,Hubei and Yunnan provinces.The chromatographic conditions were Hypersil ODS C18 column(4.6 mm× 200 mm,5 μm),acetonitrile-0.5%phosphoric acid(70∶30)as mobile phase,flow rate being 0.8 mL/min,column temperature at 30 ℃,detection wavelength at 242 nm.Results Pathymic acid showed a fine linear relationship with the peak areas between 0.362~9.050 μg(r =0.9999).The average recovery is 99.54% with RSD=1.01%.The content of Pachymic acid in three localities is Hubei>Yunnan>Anhui.Conclusion The method is proved to be feasible for quality assessment of Poria.
