补阳还五汤对缺氧-复氧损伤内皮细胞的保护作用

【摘要】 目的探讨补阳还五汤(BHD)对缺氧-复氧损伤内皮细胞(ECV)的保护作用。方法体外培养人脐静脉内皮细胞株ECV304,缺氧-复氧造成ECV损伤,同时加入不同剂量BHD,检测细胞丙二醛(MDA)、培养上清液中乳酸脱氢酶(LDH)、内皮素-1(ET-1)及一氧化氮(NO)的含量变化。结果ECV304缺氧-复氧后,与空白组比较,LDH、MDA及ET-1含量显著升高(P<0.01),而NO含量显著下降(P<0.05)。当加入低、中、高剂量的BHD或尼莫地平阳性对照药后,与缺氧-复氧模型组比较,LDH、MDA及ET-1含量下降(P<0.01或P<0.05),而NO含量升高(P<0.01)。结论BHD能抑制缺氧-复氧对ECV的损伤,对缺氧-复氧环境下的ECV起保护作用。

【Abstract】 Objective To investigate protective effects of Buyang Huanwu decoction (BDH) on endothelial cell line ECV304 cultured in hypoxia reoxygenation circumstance. Methods The effects of ECV304 were determined by measuring MDA in cells and LDH, ET-1, NO in the supernatant. Results In the circumstance of hypoxia reoxygenation, the contents of LDH, MDA, ET-1 were markedly increased (P<0.01), and the contents of NO were markedly reduced (P<0.05). Treatment with different concentrations of BHD or controlling drug either caused a reduction in the LDH, MDA, ET-1 contents and an increase in the NO contents (P<0.01 or P<0.05). Conclusions BHD can inhibit the release of LDH, MDA, ET-1 and promote the release of NO in endothelial cells in the circumstance of hypoxia reoxygenation. In this way, it may be effective in the protection of endothelial cells in the circumstance of hypoxia reoxygenation.
