【摘要】 目的研究膀胱移行细胞癌(TCC)组织中多药耐药基因MDR-1与P63蛋白及Survivin基因表达的关系,从分子水平探讨预测化疗效果的可行性。方法利用免疫组织化学方法检测10例正常膀胱粘膜标本(正常膀胱组),46例未经任何化疗的TCC标本(未化疗组),20例化疗后多次复发而行手术切除的TCC标本(获得性耐药组)组织中MDR-1、P63和Survivin的表达。结果(1)MDR-1的阳性表达在获得性耐药组与未化疗组之间有显著差异性;(2)P63的阳性表达在未化疗组各级(G)组间有显著差异;在获得性耐药组与未化疗组之间有显著差异;(3)Survivin的阳性表达在各级(G)组间有显著差异,各期(T)组间有显著差异,获得性耐药组与未化疗组间有显著差异。(4)Survivin的阳性表达率和肿瘤分级、临床分期呈正相关。结论通过检测MDR-1基因可以对TCC患者进行化学治疗敏感性预测;MDR-1表达与P63和Survivin的表达具有一致性,P63、Survivin与MDR-1三者基因可能共同表达协同作用,产生更强的耐药能力。更多还原

【Abstract】 Objective To correlate the expression of P63 and Survivin to the expression of MDR-1 gene in transitional cyst carcinoma(TCC),and to evaluate the feasibility of predicting the response to chemotherapy with molecular indexs.Methods The expressions of MDR-1,P63 and Survivin were examined by means of S-P immunohistochemical technique in 10 cases of normal bladder tissues(normal group),46 cases of TCC which were not chemotherapied (no chemotherapy TCC group),20 cases of TCC which were resected after many chemotherapies (obtained MDR group).Results (1)The MDR-1 positive expression in obtained MDR group was different from no chemotherapy TCC group.(2)In no chemotherapy TCC group,there was markable statistical significance among grade groups of P63.There was markable statistical significance between obtained MDR group and no chemotherapy group.(3)In no chemotherapy TCC group,there was markable statisticas significance among grade in groups and time groups of Survivin,respectively,and between obtained MDR group and no chemotherapy group.(4)Survivin had positive correlation to clinic tumor grade and time respectively.Conclusion MDR-1 provides a useful marker to predict chemotherapy sensitivity;the expression of MDR-1 gene is correlated with the expression of P63 and Survivin,they may be working cooperatively to make more much drug resistance.更多还原
