三七总皂甙对大鼠肝脏缺血再灌注损伤中肝细胞线粒体的保护作用
Protection for rat liver mitochondria by total saponins of panax notoginseng during ischemia-reperfusion
【摘要】 目的 观察三七总皂甙(PNS)对大鼠肝脏缺血再灌注(IR)损伤中对线粒体的保护作用。方法 制作IR模型,阻断血运前将PNS经直接通道注入并保留于肝脏组织,并提取肝细胞线粒体,测定线粒体成分的超氧化物歧化酶(SOD)总活力、丙二醛(MDA)含量;观察电镜下的组织学改变;动态性对比观察再灌注早期上述指标的变化。结果 在再灌注90 min时实验组的SOD总活力明显低于相对应的对照组(P<0 .0 1) ,MDA含量明显低于相对应的对照组(P<0 .0 1) ,电镜观察表明实验组在再灌注90 m in时线粒体等超微结构的破坏明显比对照组轻。结论 PNS在大鼠肝脏IR早期主要通过直接清除自由基及提高线粒体SOD活力而减轻和预防大鼠IR肝细胞线粒体的损伤。
【Abstract】 Objective To observe the protection of total saponins of panax notoginseng(PNS) during ischemia- reperfusion(IR).Methods The IR model of rat was established as reported method,the PNS was infused and reserved in liver of rat through direct channel before blood blocked,and then the hepatocellular mitochondria was collected.The activity of superoxide dismutase (SOD) and malondial dehyde (MDA) content were determined,and the histologic changes of liver was observed by electron microscopy.Results After 90-minute reperfusion,the total activity of SOD in the PNS group (10.767±1.638nU·mg -1 .pro) was significantly lower than that in control group (15.945±2.891nU·mg -1 .pro)(P<0.01) and the content of MDA (1.822±0.240nM·mg -1 .pro) in the PNS group also was lower than that in control group (2.893±0.782nM·mg -1 .pro)(P<0.01).More mitochondria injury was observed in PNS group than that in control group.Conclusion The PNS can protect cat hepatocyte from injury by directly eliminating oxygen free radicals and boosting the activity of SOD,especially during the early IR stage.
