水稻线粒体丝氨酸羟甲基转移酶基因的电子克隆
In silico cloning of mSHMT gene from rice(Oryza sativa L.)
【摘要】 采用基于EST的电子克隆方法得到了一段长 16 11bp的cDNA序列 ,以此为信息探针搜索HTGs数据库 ,找到一个与之高度匹配的基因组DNA序列———OSJNBa0 0 5 7G0 7克隆。用FGENESH分析该克隆中的联配区域得到一个包含 14个外显子和 13个内含子的基因。该基因位于水稻第 3染色体物理图谱的 136 .0~ 137.6cM区域。推导的ORF编码 4 98个氨基酸 ,经BLASTP搜索SWISS -PROT数据库和蛋白序列的亚细胞定位显示 ,该基因编码水稻的线粒体丝氨酸羟甲基转移酶 (mSHMT)。该基因受到EST序列的完全支持 ,其中不乏来自盐胁迫、稻瘟病菌侵染等逆境处理的EST序列 ,推测该基因与水稻对逆境的应答反应有关。
【Abstract】 Using in silico cloning method based on EST seque nces,a fragment of cDNA sequence being 1611bp in length was obtained.When using this cDNA sequence as a query to search the HTG database,we found that it was highly aligned with a genomic DNA sequence that was referred as OSJNBa0057G07 clone.The result of gene prediction by employing FgeneSH showed that this gene contained 14 exons and 13 introns that was mapped to the region ranging from 136.0 cM to 137.6 cM onto the rice physical mapping of the third chromosome.The putative ORF (498 amino acids) was defined as rice mitochondrial serine hydroxymethyltransferase (mSHMT) according to the results of sequence alignment and subcellular location analysis.Notably,this gene was completely supported by EST sequences that were derived from salt-stress,blast infection,etc,which suggested that it might be responsive to a variety of biological and non-biological stress signals in rice.
