六张水稻遗传连锁图谱的比较分析
Comparison of six rice genetic linkage maps
【摘要】 利用4个水稻品种培矮64S、9311、广陆矮4号和日本晴(Nipponbare)进行杂交组合,获得6个F2群体(PA64S/9311、PA64S/Nipponbare、PA64S/GLA9、311/GLA、Nipponbare/GLA和Nipponbare/9311)。除PA64S/Nipponbare群体为180个单株外,其余5个群体均为90个单株。在成功开发出新SSR标记的基础上,合成756对SSR引物对4个亲本进行了多态性检测,并随机选用一些具有多态性的标记对F2群体进行了多态性分析,采用MAPMAKER/EXP 3.0软件构建了6张微卫星连锁图谱。覆盖全基因组的长度为825.6~2455.2 cM,定位的标记位点数为54~152个,标记间的平均遗传距离为11.81~16.15 cM。比较分析结果表明,6张图谱间及与Temnykh等绘制的图谱在标记的染色体分布和线性排列上具有较高的一致性。选用测序品种作为构图亲本有助于解析水稻基因组遗传信息,而开展水稻亚种间比较作图研究则可以促进整个禾本科植物比较基因组研究的发展。
【Abstract】 Six F2 populations were derived from four sequenced rice varieties between PA64S,9311,Nipponbare and GLA,they were PA64S/ 9311 F2,PA64S/ Nipponbare F2,PA64S/ GLA F2,Nipponbare/ GLA F2,Nipponbare/ 9311 F2 and 9311/ GLA F2.Each F2 population had 90 lines except for PA64S/ Nipponbare(180 lines).Based on the successful development of rice new microsatellite markers,756 pairs of simple sequence repeats(SSRs) primers were synthesized in this research to analyze their polymorphisms between the four parents,then randomly selected part of polymorphic SSRs markers to analyze their F2 lines,finally constructed six microsatellite linkage maps.On these six maps,the covering length of total genome was from 825.6 cM to 2455.2 cM,the number of markers was from 54 to 152,and the average genetic distance between two markers was from 11.81 cM to 16.15 cM.In addition,99 new SSRs markers were anchored on these six genetic maps,which developed by our laboratory.The comparison between the six microsatellite genetic maps, and their comparison with the map of IR64/ Azucena which constructed by Temnykh et al had been done,results indicated that all six maps constructed in this research had high similarity of marker linear alignment to the map of IR64/ Azucena. Choosing sequenced rice varieties to construct genetic map and comparison among rice subspecies were very useful for the annotation of rice genomic genetic informations,and could facilitate the development of whole Poaceae comparative research.
