Apelin激活大鼠主动脉一氧化氮系统的研究
Apelin Activates Nitric Oxide Pathway in Isolated Aorta in Rat
【摘要】 目的研究Apelin对大鼠主动脉L-精氨酸转运、一氧化氮合酶(NOS)活性和NO生成的影响。方法体外血管薄片孵育,测定孵育液中亚硝酸盐(NO2-)含量、L-精氨酸转运及血管NOS活性。结果大鼠主动脉孵育3h后,Apelin(10-9~10-7mol/L)呈浓度依赖地刺激血管NO2-生成增加(33%、46%和69%,与对照组比较,均P<0.01);血管总NOS活性呈浓度依赖性(total nitric oxide sythase,tNOS)增加,分别较对照组增加0.68、2.36和2.5倍(均P<0.01),且以内皮型NOS(endothelial NOS,eNOS)活性升高(均为P<0.01)为主,较对照组分别增加2、4.6和5.5倍,而诱导型NOS(inducible NOS,iNOS)活性并无明显改变。结论Apelin通过激活血管组织NOS/NO系统,增加血管L-精氨酸转运与NO生成,引起血管的扩张。
【Abstract】 Objective To investigated the activation of apelin in the transport of L-arginine and nitric oxide synthase(NOS)/nitric oxide(NO) pathway in isolated incubated aorta in rat.Methods The aortic slices were incubated in vitro.The medium nitrite content,vascular NOS activities and L-arginine transport were measured.Results After incubation of aorta for 3 hour,nitrite production increased markedly compared with control(33%、46% and 69%,all P<0.01) in apelin dose dependent manner.The total NOS activities elevated significantly(0.68,2.36,2.5times) compared with control in apelin dose dependent manner.The endothelial NOS(eNOS) activities enhanced mainly(2,4.6,5.5times) compared with control(all P<0.01).However,the inducible NOS activities did not change(P>0.05).Conclusion These results suggest that apelin activate(NOS/NO) and link to the transport of L-arginine and the increase of NO contents and vasodilatation in aorta.
