辛芷鼻敏胶囊对变应性鼻炎大鼠IL-2和IL-4 mRNA表达的调节

【摘要】 目的:探讨新疆医科大学研制开发的中药新药辛芷鼻敏胶囊治疗大鼠变应性鼻炎的免疫调节作用机制。方法:用卵清蛋白致敏大鼠制成变应性鼻炎动物模型,用逆转录-多聚酶链反应(RT-PCR)对脾脏IL-2mRNAI、L-4mRNA进行定量检测。结果:与空白对照组比较,模型组IL-2mRNA表达下调,IL-4mRNA表达上调(P<0.01);与模型组比较,实验组IL-2mRNA表达上调,IL-4mRNA表达下调(P<0.01);与模型组比较,阳性对照组IL-2mRNA表达上调(P<0.05),但IL-4mRNA表达水平差异无统计学意义(P>0.05)。结论:辛芷鼻敏胶囊能通过抑制变应性鼻炎IL-4mRNA表达,上调IL-2mRNA的表达,调节Th1和Th2平衡,从而治疗变应性鼻炎。

【Abstract】 Objectives: To study the therapeutic mechanism of Xinzhi Bimin capsule in allergic rhinitis(AR).Methods: Ovalbumin sensitized white rats used as animal of AR were taken orally with the capsule of Xinzhi Bimin.IL-2 mRNA and IL-4mRNA in the spleen were used to do revers transcriptive polymerase chain reaction(RT-PCR).Results: Compared with the normal control group,IL-2mRNA in the spleen were down-regulated in the model group(P<0.01)and IL-4mRNA were up-regulated(P<0.01).The mean expression level of IL-2 mRNA in the experimental group were higher significantly than the model group,but that of IL-4 mRNA was lower evidently(P<0.05),while the effect of the positive control group was not significant(P>0.05).Conclusions: The expressions of IL-4mRNA is up-regulated and that of IL-2mRNA is down-regulated in the spleen of allergic rhinitis rats.The capsule of Xinzhi Bimin are suggested to be changed the excursion of Th1 and Th2,by means of regulating the expression of IL-2mRNA and IL-4mRNA.The result suggests that the capsule of Xinzhi Bimin was a selective and effective Chinese herbal to treat the allergic rhinitis.
